  • The Bovine Oncostatin M (OSM) ELISA is designed for the quantitative measurement of Oncostatin M in cell culture supernatants from cattle (Bos taurus) and can also be used to measure OSM in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as liver, lung, spleen, and lymph nodes. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring OSM in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. OSM is a cytokine primarily produced by activated T cells, monocytes, and macrophages, and it plays a central role in inflammation, hematopoiesis, tissue remodeling, and immune regulation. In the bovine animal model, OSM is widely studied in the context of immune response, inflammatory disease, mastitis, and tissue repair research, where its expression reflects immune activation and cytokine-mediated signaling pathways. Measurement of OSM provides valuable insight into immune regulation, inflammatory signaling, disease progression, and the efficacy of therapeutic or vaccine interventions, with reported levels in cattle typically detected at low picogram-per-milliliter concentrations under basal conditions (~10-150 pg/mL in serum or plasma) and increasing during immune stimulation, infection, or inflammatory challenge, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant bovine OSM protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Bovine Oncostatin M (OSM) is a multifunctional cytokine belonging to the IL-6 family of cytokines, which also includes IL-6, IL-11, leukemia inhibitory factor (LIF), cardiotrophin-1 (CT-1), ciliary neurotrophic factor (CNTF), cardiotrophin-like cytokine factor 1 (CLCF1), and IL-27. In cattle (Bos taurus), OSM signals through receptor complexes containing the shared gp130 subunit, typically pairing with OSM receptor (OSMR) or LIF receptor (LIFR), leading to activation of intracellular signaling pathways such as JAK/STAT3, MAPK, and PI3K. Bovine OSM is produced primarily by activated T cells, macrophages, and neutrophils and plays roles in inflammation, hematopoiesis, and tissue remodeling. In bovine health, OSM-mediated signaling contributes to regulation of immune responses during mastitis, bovine respiratory disease complex (BRDC), and other inflammatory conditions, where it can influence leukocyte activation, cytokine production, and tissue repair. OSM may also participate in fibrotic responses and regulation of epithelial and stromal cell function during chronic inflammation. As a member of the IL-6 cytokine family involved in immune regulation and tissue remodeling, bovine OSM is important for studies of host–pathogen interactions, inflammatory disease mechanisms, and cytokine signaling pathways relevant to cattle health and productivity.
