  • The Rabbit IL-1β (Interleukin-1 beta) ELISA is designed for the quantitative measurement of IL-1β in cell culture supernatants from rabbits (Oryctolagus cuniculus) and can also be used to measure IL-1β in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, liver, lung, kidney, and synovial tissue. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-1β in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-1β is a potent pro-inflammatory cytokine produced primarily by activated macrophages, monocytes, and dendritic cells and plays a central role in innate immune activation, fever induction, leukocyte recruitment, and tissue inflammation. In the rabbit animal model, IL-1β is widely studied in the context of infectious disease, inflammatory disorders, arthritis, cardiovascular research, and vaccine response, where its expression reflects activation of inflammasome-mediated immune pathways. Measurement of IL-1β provides valuable insight into inflammatory signaling, disease progression, host-pathogen interactions, and the efficacy of anti-inflammatory or immunomodulatory interventions, with reported levels in rabbits typically detected at low picogram-per-milliliter concentrations under basal conditions (~5-80 pg/mL in serum or plasma) and increasing during infection, tissue injury, or inflammatory stimulation, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant rabbit IL-1β protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Rabbit Interleukin-1 Beta (IL-1β) is a potent pro-inflammatory cytokine produced primarily by activated macrophages, monocytes, and dendritic cells in rabbits (Oryctolagus cuniculus) in response to infection, tissue injury, or inflammatory stimuli. Synthesized as an inactive precursor (pro–IL-1β), it is processed into its active form by caspase-1 within inflammasome complexes—most notably the NLRP3 inflammasome—and subsequently signals through the IL-1 receptor type 1 (IL-1R1), activating NF-κB and MAPK pathways that drive fever, leukocyte recruitment, endothelial activation, and amplification of inflammatory responses. In rabbit health, IL-1β is elevated during bacterial infections such as Pasteurella multocida, viral diseases including rabbit hemorrhagic disease virus (RHDV) and myxoma virus, and inflammatory conditions affecting the respiratory tract, joints, and gastrointestinal system. Excessive IL-1β production can contribute to systemic inflammation and tissue pathology. In research settings, rabbits serve as important models for atherosclerosis, osteoarthritis, ophthalmic inflammation, and infectious disease, and characterization of rabbit IL-1β supports studies of inflammasome activation, cytokine-mediated tissue injury, and evaluation of anti-inflammatory and immunomodulatory therapies relevant to both veterinary medicine and human inflammatory disease research.
