  • The Swine Oncostatin M (OSM) ELISA is designed for the quantitative measurement of Oncostatin M in cell culture supernatants from pigs (Sus scrofa) and can also be used to measure OSM in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as lung, liver, spleen, lymph nodes, and joint tissue. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring OSM in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. Oncostatin M is a pleiotropic cytokine produced primarily by activated macrophages, neutrophils, T cells, and stromal cells and is a member of the IL-6 cytokine family, playing a key role in inflammation, tissue remodeling, fibrosis, hematopoiesis, and bone metabolism. In the swine animal model, Oncostatin M is increasingly studied in the context of inflammatory disease, respiratory and joint disorders, immune regulation, and translational biomedical research, where its expression reflects inflammatory signaling and tissue remodeling processes. Measurement of Oncostatin M provides valuable insight into cytokine signaling pathways, disease mechanisms, and the evaluation of anti-inflammatory or anti-fibrotic therapeutic strategies, with reported levels in pigs typically detected at low picogram-per-milliliter concentrations under basal conditions (~10-120 pg/mL in serum or plasma) and increasing during immune activation, tissue injury, or disease progression, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant swine Oncostatin M protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Swine Oncostatin M (OSM) is a multifunctional cytokine belonging to the IL-6 cytokine family, which also includes IL-6, IL-11, leukemia inhibitory factor (LIF), cardiotrophin-1 (CT-1), ciliary neurotrophic factor (CNTF), cardiotrophin-like cytokine factor 1 (CLCF1), and IL-27. In pigs (Sus scrofa domesticus), OSM signals through receptor complexes containing the shared gp130 (IL6ST) subunit together with the oncostatin M receptor (OSMR), activating intracellular signaling pathways including JAK/STAT (particularly STAT3), MAPK, and PI3K/AKT. Swine OSM is produced primarily by activated T lymphocytes, macrophages, neutrophils, and dendritic cells and functions in regulation of inflammation, immune responses, hematopoiesis, and tissue remodeling. In swine health, OSM-mediated signaling is relevant during respiratory and systemic infections, including diseases such as porcine reproductive and respiratory syndrome virus (PRRSV) and swine influenza, where cytokine-driven inflammatory responses influence leukocyte activation and tissue repair in affected organs. OSM may also contribute to fibrotic responses and epithelial or stromal cell regulation during chronic inflammation. Because pigs are widely used as large-animal models for human inflammatory and metabolic diseases, characterization of swine OSM supports research into IL-6 family cytokine signaling, host–pathogen interactions, inflammatory disease mechanisms, and tissue remodeling, with applications in both veterinary medicine and translational biomedical research.
