  • The Ferret IFN-γ (Interferon gamma) ELISA is designed for the quantitative measurement of IFN-γ in cell culture supernatants from ferrets (Mustela putorius furo) and can also be used to measure IFN-γ in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as lung, spleen, lymph nodes, and peripheral blood mononuclear cells. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IFN-γ in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IFN-γ is a key type II interferon produced primarily by activated T cells and natural killer cells and plays a central role in cell-mediated immunity, antiviral defense, macrophage activation, and Th1 immune responses. In the ferret animal model, IFN-γ is widely studied in the context of influenza and other respiratory infections, vaccine evaluation, immune response characterization, and translational infectious disease research, where its expression reflects T cell-mediated immune activation. Measurement of IFN-γ provides valuable insight into cell-mediated immune responses, cytokine signaling, disease progression, and the efficacy of vaccines or immunomodulatory interventions, with reported levels in ferrets typically detected at low picogram-per-milliliter concentrations under basal conditions (~10-150 pg/mL in serum or plasma) and increasing during infection, vaccination, or immune stimulation, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant ferret IFN-γ protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Ferret Interferon-Gamma (IFN-γ) is a type II interferon produced primarily by activated CD4⁺ and CD8⁺ T lymphocytes and natural killer (NK) cells in ferrets (Mustela putorius furo), where it plays a central role in cell-mediated (Th1-type) immunity. IFN-γ signals through the IFN-γ receptor complex (IFNGR1/IFNGR2), activating JAK/STAT1 pathways and inducing genes involved in macrophage activation, enhanced antigen presentation via upregulation of MHC class I and II molecules, and promotion of cytotoxic T-cell responses. In ferret health and research, IFN-γ is particularly important in models of respiratory viral infections such as influenza virus and SARS-CoV-2, where Th1 responses contribute to viral clearance but may also influence pulmonary inflammation and immunopathology. IFN-γ is also relevant in studies of bacterial infections, vaccine-induced immunity, and T-cell–mediated protection.
