  • The Human IL-17A (Interleukin-17A) ELISA is designed for the quantitative measurement of IL-17A in cell culture supernatants from humans (Homo sapiens) and can also be used to measure IL-17A in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as lung, gut, skin, and lymphoid tissues. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring IL-17A in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. IL-17A is a pro-inflammatory cytokine produced primarily by Th17 cells, γδ T cells, and innate lymphoid cells and plays a central role in neutrophil recruitment, mucosal immunity, inflammation, and host defense against bacterial and fungal pathogens. In human studies and translational research, IL-17A is widely investigated in the context of autoimmune disease, infectious disease, inflammatory disorders, and vaccine response, where its expression reflects Th17-mediated immune activation. Measurement of IL-17A provides valuable insight into immune signaling, inflammatory pathways, disease progression, and the evaluation of immunomodulatory or therapeutic interventions, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions (~5-150 pg/mL in serum or plasma) and increasing during immune stimulation, infection, or inflammatory challenge, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant human IL-17A protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Human IL-17A (Interleukin-17A) is a pro-inflammatory cytokine belonging to the IL-17 family that plays a central role in host defense and immune-mediated inflammation. IL-17A is produced primarily by Th17 CD4⁺ T cells, as well as by γδ T cells, innate lymphoid cells (ILC3s), and other activated lymphocyte subsets in response to microbial infection and inflammatory stimuli. It acts on epithelial, endothelial, and stromal cells to induce the expression of pro-inflammatory cytokines (e.g., IL-6, TNF-α), chemokines (such as CXCL8/IL-8), and antimicrobial peptides, thereby promoting neutrophil recruitment and enhancing mucosal and barrier immunity. In healthy individuals, basal IL-17A levels are typically low, but concentrations increase during bacterial and fungal infections, particularly at mucosal surfaces. Dysregulated IL-17A signaling has been strongly implicated in chronic inflammatory and autoimmune diseases, including psoriasis, psoriatic arthritis, ankylosing spondylitis, rheumatoid arthritis, inflammatory bowel disease, and multiple sclerosis. Targeted inhibition of IL-17A (e.g., with monoclonal antibodies such as secukinumab and ixekizumab) has demonstrated significant clinical efficacy in several of these conditions. In clinical and translational research, human IL-17A serves as a key biomarker of Th17-mediated immune activation and neutrophilic inflammation and represents an important therapeutic target in immune-mediated inflammatory disease.
