  • The Human LIF (Leukemia Inhibitory Factor) ELISA is designed for the quantitative measurement of LIF in cell culture supernatants from humans (Homo sapiens) and can also be used to measure LIF in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as uterus, placenta, liver, bone marrow, and lung. While the kit is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring LIF in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. LIF is a pleiotropic cytokine produced primarily by stromal cells, macrophages, endothelial cells, and trophoblasts and plays a central role in stem cell maintenance, hematopoiesis, implantation, inflammation, and tissue regeneration. In human and non-human primate animal models, LIF is widely investigated in the context of reproductive biology, stem cell biology, tissue repair, oncology, and immune regulation, where its expression reflects critical signaling pathways in cellular proliferation and differentiation. This assay can also measure LIF from Gorilla gorilla gorilla (western lowland gorilla), Hylobates moloch (silvery gibbon), Nomascus leucogenys (northern white-cheeked gibbon), Pan paniscus (pygmy chimpanzee), Pan troglodytes (chimpanzee), and Pongo abelii (Sumatran orangutan), highlighting its utility for comparative and translational studies across closely related primate species due to the high sequence conservation of LIF. Human Leukemia Inhibitory Factor (LIF) is a multifunctional cytokine belonging to the IL-6 family—which includes IL-6, IL-11, oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), cardiotrophin-like cytokine factor 1 (CLCF1), and IL-27—all of which signal through receptor complexes containing the shared gp130 subunit. LIF binds to the LIF receptor (LIFR) in association with gp130, activating JAK/STAT3, MAPK, and PI3K signaling pathways that regulate cell survival, differentiation, immune modulation, and tissue repair. In humans, LIF plays critical roles in embryonic implantation and endometrial receptivity, hematopoiesis, neural development, and maintenance of stem cell pluripotency. LIF-mediated STAT3 signaling is also involved in modulation of inflammatory responses and tissue regeneration; however, dysregulated LIF expression has been implicated in fibrosis, chronic inflammatory diseases, and tumor progression in cancers such as breast, pancreatic, and colorectal cancer, where it can promote tumor cell survival and stromal remodeling. Because of its dual roles in regeneration and pathology, human LIF is an important therapeutic target and biomarker in reproductive medicine, oncology, and fibrotic disease research. Characterization of LIF signaling continues to inform development of STAT3-modulating therapies and strategies aimed at balancing tissue repair with control of chronic inflammation and malignancy.
