  • The Dolphin LIF (Leukemia Inhibitory Factor) ELISA is designed for the quantitative measurement of LIF in cell culture supernatants from dolphins (family Delphinidae) and can also be used to measure LIF in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as lung, liver, spleen, placenta, and reproductive tissues. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring LIF in other sample types to ensure optimal sensitivity and accuracy, allowing adjustment of buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. LIF is a pleiotropic cytokine and a member of the IL-6 cytokine family, produced by stromal cells, macrophages, endothelial cells, and epithelial cells, and plays an important role in immune regulation, tissue repair, hematopoiesis, inflammation, and reproductive biology. In dolphin animal health research, LIF is increasingly studied as a marker of immune status, inflammatory responses, tissue regeneration, and reproductive health, particularly in the context of infectious disease, environmental stressors, pollutant exposure, and overall population health monitoring. Dolphin Leukemia Inhibitory Factor (LIF) is a multifunctional cytokine belonging to the IL-6 family—which includes IL-6, IL-11, oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), cardiotrophin-like cytokine factor 1 (CLCF1), and IL-27—all of which signal through receptor complexes containing the shared gp130 subunit. In dolphins, particularly the bottlenose dolphin (Tursiops truncatus), LIF binds to the LIF receptor (LIFR) in association with gp130, activating JAK/STAT3, MAPK, and PI3K signaling pathways that regulate cell survival, differentiation, immune modulation, and tissue repair. Dolphin LIF is likely involved in reproductive processes such as embryo implantation and placental development, as well as in hematopoiesis and neural tissue maintenance.
