  • The Mouse GM-CSF (Granulocyte-Macrophage Colony-Stimulating Factor) ELISA is designed for the quantitative measurement of GM-CSF in cell culture supernatants from mice (Mus musculus) and can also be used to measure GM-CSF in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, bronchoalveolar lavage fluid, and tissue lysates from organs such as lung, spleen, lymph nodes, and bone marrow. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring GM-CSF in other sample types to ensure optimal sensitivity and accuracy, allowing adjustment of buffers, antibody concentrations, incubation times, temperatures, and other assay parameters as needed. GM-CSF is a cytokine produced primarily by activated T cells, macrophages, endothelial cells, and fibroblasts and plays a critical role in hematopoiesis, myeloid cell differentiation, dendritic cell development, and inflammatory immune responses. In the mouse animal model, GM-CSF is extensively studied in the context of pulmonary inflammation, autoimmune disease, infection, cancer, and vaccine response, where its expression reflects activation of innate and adaptive immune pathways. Measurement of mouse GM-CSF provides valuable insight into immune signaling, inflammatory cascades, disease mechanisms, and therapeutic intervention outcomes, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions (~5-150 pg/mL) and increasing during immune stimulation, infection, or inflammatory challenge, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant mouse GM-CSF protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Mouse GM-CSF (Granulocyte–Macrophage Colony-Stimulating Factor, CSF2) is a hematopoietic cytokine belonging to the colony-stimulating factor family, which also includes G-CSF (CSF3) and M-CSF (CSF1) that regulate the development, proliferation, and activation of myeloid immune cells. In mice (Mus musculus), GM-CSF is produced by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to infection, inflammatory cytokines, or immune stimulation. Mouse GM-CSF binds to the GM-CSF receptor (CSF2R) expressed on hematopoietic progenitor cells and mature myeloid cells, activating intracellular signaling pathways such as JAK/STAT, MAPK/ERK, and PI3K–AKT, which promote the proliferation and differentiation of granulocytes, macrophages, and dendritic cells, as well as enhance their immune effector and antigen-presenting functions. In murine immunity, GM-CSF plays a critical role in regulating inflammatory responses and development of antigen-presenting cells. Mice are widely used as animal models for studying immune regulation and inflammatory disease, and GM-CSF has been extensively studied in mouse models of autoimmune diseases such as experimental autoimmune encephalomyelitis (EAE), a widely used animal model for human multiple sclerosis, where GM-CSF produced by T cells drives pathogenic myeloid cell activation and central nervous system inflammation, providing key insights into immune-mediated neuroinflammatory disease mechanisms.
