  • The Bovine GM-CSF (Granulocyte-Macrophage Colony-Stimulating Factor) ELISA is designed for the quantitative measurement of GM-CSF in cell culture supernatants from cattle (Bos taurus) and can also be used to measure GM-CSF in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, milk, and tissue lysates from organs such as lung, liver, spleen, lymph nodes, and bone marrow. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring GM-CSF in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. GM-CSF is a cytokine produced primarily by activated T cells, macrophages, endothelial cells, fibroblasts, and epithelial cells and plays a critical role in hematopoiesis, myeloid cell differentiation, dendritic cell development, and inflammatory immune responses. In the bovine animal model, GM-CSF is widely studied in the context of infectious disease, mastitis, respiratory disease, vaccine development, and immune modulation, where its expression reflects activation of innate and adaptive immune pathways important for animal health and productivity. Measurement of bovine GM-CSF provides valuable insight into immune signaling, inflammatory cascades, disease progression, and the evaluation of vaccines or immunotherapeutic interventions, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions and increasing during infection, inflammation, or immune stimulation, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant bovine GM-CSF protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Bovine GM-CSF (Granulocyte–Macrophage Colony-Stimulating Factor, CSF2) is a hematopoietic cytokine belonging to the colony-stimulating factor family, which also includes G-CSF (CSF3) and M-CSF (CSF1) that regulate the development, proliferation, and activation of myeloid immune cells. In cattle (Bos taurus), GM-CSF is produced by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and other stromal cells in response to infection, inflammatory cytokines, or immune stimulation. Bovine GM-CSF binds to the GM-CSF receptor (CSF2R) expressed on hematopoietic progenitor cells and mature myeloid cells, activating intracellular signaling pathways such as JAK/STAT, MAPK/ERK, and PI3K–AKT, which promote the proliferation and differentiation of granulocytes, macrophages, and dendritic cells, as well as enhance their functional activity in innate and adaptive immune responses. In bovine health, GM-CSF contributes to host defense against bacterial infections, particularly by regulating macrophage and neutrophil activity in tissues such as the mammary gland and respiratory tract. Bovine GM-CSF has also been studied in animal models of bovine tuberculosis caused by Mycobacterium bovis, where GM-CSF influences macrophage activation and antigen-presenting cell development, helping researchers understand cell-mediated immunity and vaccine responses to mycobacterial infections relevant to both livestock health and human tuberculosis research.
