  • The FNDC5 (Fibronectin Type III Domain-Containing Protein 5) ELISA is designed for the quantitative measurement of FNDC5 in cell culture supernatants and can also be used to measure FNDC5 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, skeletal muscle lysates, adipose tissue, brain tissue, and other metabolically active tissues. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring FNDC5 in other sample types to ensure optimal sensitivity and accuracy, allowing adjustment of buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. FNDC5 is a type I membrane protein that is proteolytically cleaved to generate irisin, a circulating myokine involved in energy metabolism, thermogenesis, glucose homeostasis, muscle-adipose tissue crosstalk, and neuroprotection. FNDC5 expression is strongly regulated by exercise and metabolic status and is widely studied in the context of metabolic disease, obesity, diabetes, cardiovascular disease, muscle physiology, and neurobiology. Importantly, the FNDC5 protein is highly conserved across many species, supporting its broad relevance in comparative biology, translational research, and animal model studies, and enabling cross-species investigation of metabolic and exercise-induced signaling pathways. Measurement of FNDC5 provides valuable insight into metabolic regulation, tissue adaptation, and systemic energy balance, with reported levels typically detected at low picogram- to nanogram-per-milliliter concentrations, depending on species, sample type, and physiological condition. The kit contains a capture antibody, detection antibody, and recombinant FNDC5 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. FNDC5 (Fibronectin Type III Domain–Containing Protein 5) is a membrane-associated protein that serves as the precursor of the myokine irisin, a hormone-like peptide involved in energy metabolism and metabolic regulation. FNDC5 belongs to a small group of proteins containing fibronectin type III domains, which are structural motifs commonly involved in cell signaling and extracellular interactions. FNDC5 is expressed primarily in skeletal muscle, but is also found in cardiac muscle, adipose tissue, and other metabolically active tissues. During exercise or metabolic stimulation, FNDC5 can be proteolytically cleaved to release irisin, which circulates in the bloodstream and promotes browning of white adipose tissue, increased thermogenesis, and improved energy expenditure. FNDC5 expression is regulated in part by PGC-1α, a transcriptional coactivator involved in mitochondrial biogenesis and metabolic adaptation to exercise. Because of its role in exercise physiology, metabolic homeostasis, and energy balance, FNDC5/irisin signaling has been widely studied in metabolic disease, obesity, diabetes, and muscle biology, making it an important molecule in endocrine and metabolic research.
