  • The Chicken M-CSF (Macrophage Colony-Stimulating Factor) ELISA is designed for the quantitative measurement of M-CSF in cell culture supernatants from chickens (Gallus gallus domesticus) and can also be used to measure M-CSF in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, liver, lung, bone marrow, and intestinal tissues. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring M-CSF in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. M-CSF is a hematopoietic growth factor produced by stromal cells, fibroblasts, endothelial cells, and epithelial cells and is essential for monocyte and macrophage differentiation, survival, and function. In the chicken animal model, M-CSF is widely studied for its role in innate immune development, macrophage biology, host-pathogen interactions, inflammatory responses, and vaccine-induced immunity, making it a key biomarker in avian immunology and poultry health research. Measurement of chicken M-CSF provides valuable insight into myeloid lineage regulation, immune activation, and disease progression, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions and increasing during infection, inflammation, or immune stimulation, depending on sample type and physiological status. The kit contains a capture antibody, detection antibody, and recombinant chicken M-CSF protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Chicken Macrophage Colony-Stimulating Factor (M-CSF) is a cytokine that regulates the development, proliferation, and survival of monocytes and macrophages in chickens (Gallus gallus). M-CSF is produced by several cell types, including fibroblasts, epithelial cells, and stromal cells, in response to tissue signals and inflammatory stimuli. Chicken M-CSF binds to the colony-stimulating factor-1 receptor (CSF1R), a receptor tyrosine kinase expressed on monocytes and macrophage lineage cells, activating intracellular signaling pathways such as PI3K–AKT, ERK/MAPK, and JAK–STAT that promote differentiation and maintenance of macrophage populations. Through these mechanisms, M-CSF supports macrophage-mediated immune surveillance, phagocytosis, tissue homeostasis, and inflammatory responses. Because macrophages play a key role in host defense, M-CSF–mediated macrophage function is important for maintaining immune health and resistance to infection in poultry. In cell culture, recombinant chicken M-CSF is commonly used to promote differentiation of chicken monocytes into macrophages and to support macrophage proliferation and functional assays in vitro. Chicken M-CSF is also studied in avian immunology and animal model research, including investigations of macrophage responses to important poultry pathogens such as avian influenza virus, Salmonella spp., and Eimeria species.
