  • The Zebrafish TNF-α (Tumor Necrosis Factor-alpha) ELISA is designed for the quantitative measurement of TNF-α in cell culture supernatants from zebrafish (Danio rerio) and can also be used to measure TNF-α in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from organs such as spleen, liver, intestine, kidney, and gill tissues. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring TNF-α in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. TNF-α is a proinflammatory cytokine produced primarily by macrophages, monocytes, neutrophils, and endothelial cells, and plays a central role in innate immune responses, inflammation, infection control, and tissue homeostasis. In the zebrafish animal model, TNF-α is widely studied in the context of host-pathogen interactions, inflammatory disease, tissue injury, toxicology, and drug development, where its expression reflects activation of innate immune pathways. Measurement of zebrafish TNF-α provides valuable insight into immune signaling, inflammatory cascades, disease progression, and the evaluation of therapeutic or immunomodulatory interventions, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions and increasing during immune challenge, infection, or inflammatory stress, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant zebrafish TNF-α protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Zebrafish (Danio rerio) Tumor Necrosis Factor Alpha (TNF-α) is a proinflammatory cytokine that plays an important role in innate immune responses, inflammation, and host defense against pathogens in teleost fish. TNF belongs to the tumor necrosis factor (TNF) superfamily, a group of immune signaling molecules that includes TNF-α, lymphotoxins, Fas ligand (FasL), CD40 ligand (CD40L), and TRAIL, which regulate inflammatory signaling, apoptosis, and immune cell activation. In zebrafish, TNF is primarily produced by activated macrophages, neutrophils, and other innate immune cells following infection, tissue injury, or recognition of pathogen-associated molecular patterns. TNF signals through TNF receptor homologs that activate intracellular pathways such as NF-κB and MAPK, leading to induction of proinflammatory cytokines, antimicrobial responses, and recruitment of immune cells to sites of infection or injury. Zebrafish TNF plays important roles in defense against bacterial and mycobacterial infections, and it has been extensively studied in zebrafish models of Mycobacterium marinum infection, which serve as a model for human tuberculosis, where TNF signaling regulates macrophage activation and granuloma formation. Because zebrafish are widely used as a genetic and imaging model for immunology and infectious disease, characterization of zebrafish TNF has provided important insights into host–pathogen interactions, inflammatory signaling, and innate immune responses relevant to vertebrate immunity and human disease.
