  • The Atlantic Salmon TNF-α (Tumor Necrosis Factor-alpha) ELISA is designed for the quantitative measurement of TNF-α in cell culture supernatants from Atlantic salmon (Salmo salar) and can also be used to measure TNF-α in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, and tissue lysates from immune- and disease-relevant tissues such as head kidney, spleen, liver, intestine, gill, and skin. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring TNF-α in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay parameters as needed. TNF-α is a key proinflammatory cytokine produced primarily by macrophages, monocytes, neutrophils, and other innate immune cells and plays a central role in inflammatory signaling, host defense against pathogens, apoptosis, and immune regulation. In the Atlantic salmon animal model, TNF-α is widely studied in the context of bacterial and viral infections, parasitic disease, vaccination response, stress physiology, and aquaculture health management, where its expression reflects activation of innate immune pathways critical for fish health, disease resistance, and production performance. Measurement of Atlantic salmon TNF-α provides valuable insight into immune activation, inflammatory cascades, disease progression, and the evaluation of vaccines or immunomodulatory treatments, with reported levels typically detected at low picogram-per-milliliter concentrations under basal conditions and increasing during infection, inflammation, or environmental stress, depending on sample type and experimental context. The kit contains a capture antibody, detection antibody, and recombinant Atlantic salmon TNF-α protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Atlantic Salmon (Salmo salar) Tumor Necrosis Factor Alpha (TNF-α) is a proinflammatory cytokine that plays an important role in innate immune responses, inflammation, and host defense against pathogens in teleost fish. TNF-α belongs to the tumor necrosis factor (TNF) superfamily, which in vertebrates includes molecules such as TNF-α, TNF-β (lymphotoxin), Fas ligand (FasL), CD40 ligand (CD40L), and TRAIL, all involved in regulating immune signaling, apoptosis, and inflammatory responses. In Atlantic salmon, TNF-α is primarily produced by activated macrophages, monocytes, and other immune cells following infection, tissue injury, or exposure to pathogen-associated molecular patterns. TNF-α signals through TNF receptor homologs that activate intracellular pathways such as NF-κB and MAPK, leading to the induction of proinflammatory cytokines, antimicrobial responses, and immune cell recruitment. In salmon, TNF-α plays a critical role in defense against bacterial and viral pathogens common in aquaculture, including Aeromonas salmonicida, Vibrio species, and infectious salmon anemia virus (ISAV), by promoting inflammatory responses and activation of innate immune defenses. TNF-α is also involved in regulating immune responses in key mucosal tissues such as the gills, skin, and intestine, which serve as major pathogen entry points in aquatic environments. Because Atlantic salmon is an important aquaculture species, characterization of salmon TNF-α supports research on fish immunology, host–pathogen interactions, and disease resistance, contributing to improved understanding of immune regulation and health management in aquaculture systems.
