  • The Swine SCF (Stem Cell Factor) ELISA is designed for the quantitative measurement of SCF in cell culture supernatants from swine (Sus scrofa) and can also be used to measure SCF in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, bone marrow aspirates, and tissue lysates from organs such as spleen, skin, lung, and gastrointestinal tissues. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring SCF in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. SCF, also known as KIT ligand, is a hematopoietic growth factor produced by stromal cells, fibroblasts, endothelial cells, and epithelial cells and plays a central role in hematopoietic stem cell maintenance, mast cell development, myeloid lineage differentiation, and immune cell survival through signaling via the c-KIT (CD117) receptor. In the swine animal model, SCF is widely studied in the context of hematopoiesis, immune development, inflammatory disease, regenerative medicine, and translational models of human health, making it a valuable biomarker in both agricultural and biomedical research. Measurement of swine SCF provides insight into stem cell signaling, immune system function, and disease progression, with reported concentrations typically detected at low to mid-picogram-per-milliliter levels in serum or plasma under basal conditions and increasing to higher picogram-per-milliliter ranges in cell culture supernatants, bone marrow samples, or disease-associated states, depending on sample type and physiological context. The kit contains a capture antibody, detection antibody, and recombinant swine SCF protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Swine Stem Cell Factor (SCF) (also known as KIT ligand, KITLG, mast cell growth factor, or steel factor) is a cytokine and growth factor that plays an important role in hematopoiesis, stem cell maintenance, and immune cell development in pigs (Sus scrofa domesticus). SCF belongs to a group of hematopoietic growth factors that regulate the survival, proliferation, and differentiation of stem and progenitor cells. It functions by binding to the c-Kit receptor (CD117), a receptor tyrosine kinase expressed on hematopoietic stem cells, mast cells, melanocytes, and germ cells, activating intracellular signaling pathways including PI3K/AKT, MAPK, and JAK/STAT that promote cell survival, proliferation, and differentiation. In pigs, SCF is produced by bone marrow stromal cells, fibroblasts, endothelial cells, and epithelial cells, and exists in both membrane-bound and soluble forms, allowing both localized and systemic regulation of c-Kit–expressing cells. Swine SCF is important for maintenance of hematopoietic stem cells within the bone marrow niche, as well as for mast cell development, pigmentation biology, and germ cell survival. Because pigs are widely used as large-animal models for human hematopoiesis, immunology, and regenerative medicine, characterization of swine SCF supports studies of stem cell regulation, immune cell development, and c-Kit–mediated signaling pathways relevant to both livestock health and translational biomedical research.
