  • The Rabbit LIF (Leukemia Inhibitory Factor) ELISA is designed for the quantitative measurement of LIF in cell culture supernatants from rabbits (Oryctolagus cuniculus) and can also be used to measure LIF in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, uterine flushings, and tissue lysates from organs such as uterus, placenta, spleen, lung, and liver. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring LIF in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. LIF is a multifunctional cytokine and a member of the IL-6 cytokine family, produced by fibroblasts, epithelial cells, endothelial cells, macrophages, and reproductive tissues, and signals through the LIF receptor (LIFR) in complex with gp130 to regulate cell differentiation, survival, inflammation, and tissue remodeling. In the rabbit animal model, LIF is especially important in studies of reproductive biology, embryo implantation, placental development, stem cell maintenance, and immune regulation, as well as inflammatory and regenerative processes. Measurement of rabbit LIF provides valuable insight into cytokine signaling, reproductive success, immune responses, and disease progression, with reported concentrations typically detected at low picogram-per-milliliter levels under basal conditions and increasing during pregnancy, tissue injury, inflammation, or immune stimulation, depending on sample type and physiological context. The kit contains a capture antibody, detection antibody, and recombinant rabbit LIF protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Rabbit Leukemia Inhibitory Factor (LIF) is a multifunctional cytokine belonging to the IL-6 family—which includes IL-6, IL-11, oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), cardiotrophin-like cytokine factor 1 (CLCF1), and IL-27—and signals through receptor complexes containing the shared gp130 subunit. In rabbits (Oryctolagus cuniculus), LIF binds to the LIF receptor (LIFR) in association with gp130, activating JAK/STAT3, MAPK, and PI3K pathways that regulate cell survival, differentiation, immune modulation, and tissue repair. Rabbit LIF plays important roles in reproductive biology, particularly in uterine receptivity and embryo implantation, and has been studied in the context of early embryonic development and stem cell maintenance. In disease models, LIF-mediated STAT3 signaling may influence inflammatory responses and tissue remodeling in experimental systems of atherosclerosis, osteoarthritis, ocular inflammation, pulmonary injury, and cancer. Dysregulated LIF expression could contribute to fibrosis or tumor progression through persistent STAT3 activation. Because rabbits are widely used as translational models in cardiovascular, musculoskeletal, and ophthalmic research, characterization of rabbit LIF supports investigations into IL-6 family cytokine signaling, regenerative biology, host–pathogen interactions, and development of STAT3-modulating therapeutic strategies relevant to human inflammatory and fibrotic diseases.
