  • The Chicken CCL20 (MIP-3α) ELISA is designed for the quantitative measurement of CCL20 in cell culture supernatants from chicken (Gallus gallus) and can also be used to measure CCL20 in a variety of biological sample types reported in the literature, including serum, plasma, intestinal tissue lysates, respiratory tissues, spleen, bursa of Fabricius, and mucosal secretions. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring CCL20 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. CCL20 is a chemokine produced by epithelial cells, macrophages, dendritic cells, and intestinal tissues in response to microbial stimulation and inflammatory signals, and it functions primarily through the CCR6 receptor to recruit lymphocytes, dendritic cells, and other immune effector cells to mucosal sites. In the avian animal model, CCL20 plays a critical role in mucosal immunity, gut health, respiratory defense, vaccine response, and host-pathogen interactions, particularly in the context of enteric infections, coccidiosis, Salmonella, and respiratory pathogens. Measurement of chicken CCL20 provides valuable insight into chemokine-mediated immune cell trafficking, barrier immunity, and inflammatory responses, with reported concentrations typically detected at low picogram-per-milliliter levels under basal conditions and increasing significantly during infection, vaccination, or inflammatory stimulation, depending on tissue source and experimental design. The kit contains a capture antibody, detection antibody, and recombinant chicken CCL20 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Chicken CCL20 (C-C motif chemokine ligand 20, also known as MIP-3α, macrophage inflammatory protein-3 alpha, or LARC – liver and activation-regulated chemokine) is a pro-inflammatory chemokine that plays an important role in mucosal immunity and leukocyte recruitment in chickens (Gallus gallus). CCL20 primarily signals through the CCR6 receptor, promoting chemotaxis of immune cells including immature dendritic cells, memory T cells, and certain B cell populations to sites of infection or epithelial inflammation. In poultry, CCL20 is strongly expressed in epithelial tissues such as the intestinal tract, respiratory mucosa, and skin, where it contributes to the early immune response against pathogens. It is particularly relevant in infections caused by Eimeria spp. (coccidiosis), avian influenza virus, Newcastle disease virus, and bacterial pathogens such as Salmonella and Campylobacter, where epithelial cells produce CCL20 to recruit immune cells and coordinate local inflammatory responses. While CCL20-mediated leukocyte trafficking supports pathogen clearance and mucosal defense, excessive expression may contribute to epithelial inflammation and tissue damage. As a biomarker of mucosal immune activation, chicken CCL20 is widely studied in investigations of host–pathogen interactions, intestinal immunity, and vaccine responses in poultry. Characterization of chicken CCL20 therefore supports research aimed at improving disease resistance, mucosal vaccine strategies, and overall flock health in commercial poultry systems.
