  • The Human CCL11 (Eotaxin-1) ELISA is designed for the quantitative measurement of CCL11 in cell culture supernatants from Homo sapiens and can also be used to measure CCL11 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, bronchoalveolar lavage fluid (BALF), cerebrospinal fluid (CSF), and tissue lysates. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring CCL11 in other sample types to ensure optimal sensitivity and accuracy, allowing researchers to adjust buffers, antibody concentrations, incubation times, temperatures, and other assay conditions as needed. CCL11 is a CC chemokine produced by epithelial cells, endothelial cells, fibroblasts, macrophages, and smooth muscle cells in response to inflammatory cytokines, and it signals primarily through the CCR3 receptor to mediate the recruitment and activation of eosinophils, basophils, and Th2 lymphocytes. In human research, CCL11 plays a central role in allergic inflammation, asthma, atopic dermatitis, eosinophilic disorders, parasitic infections, and chronic inflammatory diseases, and has also been investigated in the context of neuroinflammation and age-associated cognitive decline. Measurement of human CCL11 provides important insight into Th2-driven immune responses, chemokine-mediated cell trafficking, and inflammatory disease progression, with reported circulating concentrations typically detected at low to mid picogram-per-milliliter levels under basal conditions and elevated during allergic responses, airway inflammation, and systemic inflammatory states, depending on sample type and clinical context. The kit contains a capture antibody, detection antibody, and recombinant human CCL11 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Human CCL11 (C-C motif chemokine ligand 11), also known as eotaxin-1, is a member of the CC chemokine family that plays a central role in regulating eosinophil recruitment and type 2–associated immune responses. CCL11 is produced primarily by epithelial cells, fibroblasts, endothelial cells, smooth muscle cells, and activated immune cells in response to Th2-associated cytokines such as IL-4 and IL-13, as well as inflammatory stimuli. It functions predominantly through the CCR3 receptor to promote chemotaxis and activation of eosinophils, and also influences basophils and certain Th2 lymphocytes. In healthy individuals, circulating CCL11 levels are typically low, but concentrations increase during allergic inflammation, parasitic infection, and certain chronic inflammatory conditions. Elevated CCL11 has been implicated in asthma, allergic rhinitis, atopic dermatitis, eosinophilic esophagitis, inflammatory bowel disease, and some neuroinflammatory and aging-related processes. In clinical and translational research, human CCL11 serves as an important biomarker of eosinophilic inflammation and Th2-driven immune responses and represents a potential therapeutic target for allergic and eosinophil-associated diseases.
