  • The Human CCL2 (MCP-1) ELISA is designed for the quantitative measurement of CCL2 in cell culture supernatants from Homo sapiens and can also be used to measure CCL2 in a variety of biological sample types reported in the literature, including serum, plasma, whole blood, cerebrospinal fluid (CSF), synovial fluid, bronchoalveolar lavage fluid (BALF), and tissue lysates. While the assay is optimized for cell culture supernatants, minor modifications to the protocol may be required when measuring CCL2 in other sample types to ensure optimal sensitivity and matrix compatibility. CCL2 is a CC chemokine produced by monocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to inflammatory stimuli, and it signals primarily through the CCR2 receptor to mediate recruitment and activation of monocytes, memory T cells, and dendritic cells to sites of tissue injury or infection. In human biology, CCL2 plays a central role in acute and chronic inflammation, autoimmune disease, cardiovascular disease, metabolic disorders, neuroinflammation, and cancer progression, where monocyte trafficking and macrophage infiltration are key pathogenic mechanisms. Measurement of human CCL2 provides important insight into chemokine-driven immune cell migration, inflammatory burden, and disease progression, with circulating concentrations typically detected at low to mid picogram-per-milliliter levels under basal conditions and elevated during systemic inflammation, infection, tissue injury, or chronic inflammatory states, depending on clinical context and sample type. The kit contains a capture antibody, detection antibody, and recombinant human CCL2 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Human CCL2 (C-C motif chemokine ligand 2, also known as monocyte chemoattractant protein-1, MCP-1) is a pro-inflammatory chemokine that plays a critical role in recruitment and activation of monocytes, macrophages, dendritic cells, and memory T cells during immune responses. CCL2 primarily signals through the CCR2 receptor, activating intracellular pathways that promote leukocyte migration, cytokine production, and coordination of innate and adaptive immunity at sites of infection, tissue injury, or inflammation. In humans, CCL2 is produced by a variety of cells including endothelial cells, fibroblasts, epithelial cells, and immune cells in response to inflammatory stimuli such as infection or tissue damage. While CCL2-mediated recruitment of monocytes is essential for host defense and tissue repair, dysregulated or excessive expression contributes to chronic inflammatory diseases including atherosclerosis, rheumatoid arthritis, multiple sclerosis, inflammatory bowel disease, and chronic kidney disease. CCL2 also plays an important role in cancer biology, where it promotes recruitment of tumor-associated macrophages that can support tumor growth, angiogenesis, and metastasis. As a key regulator of monocyte trafficking and inflammatory signaling, human CCL2 is widely studied as both a biomarker of inflammation and a therapeutic target in autoimmune disease, cardiovascular disease, cancer, and other chronic inflammatory conditions.
