  • The Chicken CCL5 (RANTES) ELISA is designed for the quantitative measurement of CCL5 in cell culture supernatants from chicken (Gallus gallus) and can also be used to measure CCL5 in additional biological sample types reported in the literature, including serum, plasma, splenocyte culture supernatants, intestinal tissue lysates, respiratory tissues, bursa of Fabricius, and peripheral blood leukocyte cultures, although minor protocol modifications may be required to optimize assay performance in different matrices. CCL5 is a CC chemokine produced by activated T lymphocytes, macrophages, epithelial cells, and other immune cells in response to inflammatory and pathogenic stimuli, and it functions primarily through CCR1, CCR3, and CCR5 receptors to recruit and activate T cells, monocytes, and other leukocytes at sites of infection or tissue inflammation. In the avian animal model, CCL5 plays an important role in cell-mediated immunity, antiviral responses, mucosal defense, and vaccine-induced immune activation, particularly in the context of respiratory pathogens, enteric infections, and systemic inflammatory challenges relevant to poultry health and production. Measurement of chicken CCL5 provides valuable insight into chemokine-driven immune cell trafficking, Th1-type immune responses, and host-pathogen interactions, with concentrations typically detected at low picogram-per-milliliter levels under basal conditions and increasing following infection, vaccination, or mitogen stimulation, depending on tissue source and experimental design. The kit contains a capture antibody, detection antibody, and recombinant chicken CCL5 protein standard for assay development, and these antibodies have been validated to function together in an ELISA format using the provided standard. Chicken CCL5 (C–C motif chemokine ligand 5, also known as RANTES – Regulated upon Activation, Normal T cell Expressed and Secreted) is a proinflammatory chemokine involved in immune cell recruitment and inflammatory responses in chickens (Gallus gallus). CCL5 is a member of the CC chemokine family, which includes related chemokines such as CCL3, CCL4, and CCL2, all of which regulate leukocyte trafficking during immune activation. In chickens, CCL5 is produced primarily by activated T lymphocytes, macrophages, dendritic cells, and epithelial cells in response to infection, inflammatory stimuli, or tissue damage. CCL5 functions mainly by binding to chemokine receptors such as CCR1 and CCR5, triggering intracellular signaling pathways that promote chemotaxis and activation of T cells, monocytes/macrophages, and other immune cells at sites of infection or inflammation. In avian immunity, CCL5 plays a role in responses to viral, bacterial, and parasitic pathogens, including important poultry diseases such as avian influenza virus, Newcastle disease virus, and Eimeria infections, where recruitment of immune cells is critical for pathogen control. Because chickens are widely used in avian immunology and host–pathogen interaction studies, chicken CCL5 is studied in research on chemokine signaling, inflammatory responses, and vaccine-induced immunity, contributing to improved understanding of immune regulation and disease resistance in poultry.
