  • The Human CCL4 (MIP-1β) ELISA is designed for the quantitative measurement of CCL4 in cell culture supernatants from human cells and can also be used to measure CCL4 in additional biological sample types reported in the literature, including serum, plasma, peripheral blood mononuclear cell (PBMC) culture supernatants, bronchoalveolar lavage fluid, and tissue lysates, with minor protocol modifications recommended to optimize assay performance across different matrices. CCL4 is a CC chemokine produced by activated T lymphocytes, monocytes, macrophages, dendritic cells, and NK cells in response to inflammatory cytokines, microbial products, and viral infection, and it functions primarily through the CCR5 receptor to recruit and activate monocytes, cytotoxic T cells, and NK cells at sites of infection or tissue inflammation. Human CCL4 plays a central role in antiviral immunity, inflammatory signaling, and leukocyte trafficking and is widely studied in the context of HIV infection (due to its interaction with CCR5), autoimmune diseases, chronic inflammatory disorders, cancer immunology, and cytokine release syndromes. Measurement of human CCL4 provides valuable insight into Th1-type immune activation, host-pathogen interactions, and immune-mediated tissue damage, with basal concentrations typically low under healthy conditions and elevated during infection, immune stimulation, or inflammatory disease depending on clinical status and sample type. The ELISA kit contains a capture antibody, detection antibody, and recombinant human CCL4 protein standard validated for use together in an optimized ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Human CCL4 (C-C motif chemokine ligand 4, also known as MIP-1β, macrophage inflammatory protein-1 beta) is a pro-inflammatory chemokine that plays a key role in immune cell recruitment and regulation of inflammatory and antiviral immune responses. CCL4 primarily signals through the CCR5 receptor, promoting chemotaxis of monocytes, macrophages, dendritic cells, natural killer (NK) cells, and activated T lymphocytes to sites of infection, inflammation, or tissue injury. In humans, CCL4 is produced by activated macrophages, dendritic cells, T lymphocytes, epithelial cells, and endothelial cells following stimulation by microbial products or inflammatory cytokines such as TNF-α and IL-1β. CCL4 contributes to host defense by coordinating leukocyte trafficking and enhancing immune activation during infections; it is also involved in viral immunity, particularly in HIV infection, where CCL4 competes with the virus for the CCR5 co-receptor, thereby inhibiting viral entry into target cells. Dysregulated CCL4 expression is associated with chronic inflammatory and autoimmune diseases, including rheumatoid arthritis, multiple sclerosis, and inflammatory bowel disease, as well as cancer, where chemokine-mediated immune cell infiltration can influence tumor microenvironment dynamics. Because of its central role in immune cell migration and CCR5-mediated signaling, human CCL4 is widely studied as a biomarker of inflammation and a potential therapeutic target in infectious disease, immune-mediated disorders, and cancer immunology.
