  • The Human IL-1 beta (IL-1β) ELISA is designed for the quantitative measurement of interleukin-1 beta (IL-1β) in human cell culture supernatants and can also be used to measure IL-1β in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, cerebrospinal fluid (CSF), and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-1β is a potent pro-inflammatory cytokine produced primarily by activated monocytes, macrophages, dendritic cells, and other innate immune cells following inflammasome activation and caspase-1-mediated cleavage of pro-IL-1β, and it plays a central role in fever induction, leukocyte recruitment, endothelial activation, and amplification of inflammatory signaling pathways. In healthy individuals, circulating IL-1β levels are typically very low or undetectable (often <1-5 pg/mL in serum or plasma), but concentrations can increase to tens or hundreds of pg/mL in response to acute infection, tissue injury, autoimmune disease, cytokine release syndromes, or severe systemic inflammation such as sepsis, depending on disease severity and biological context. In human research, IL-1β serves as a key biomarker of innate immune activation, inflammasome activity, autoinflammatory disorders, infectious disease progression, and therapeutic response to anti-inflammatory or biologic treatments. The ELISA kit contains a capture antibody, detection antibody, and recombinant human IL-1β protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Human Interleukin-1 Beta (IL-1β) is a potent pro-inflammatory cytokine produced primarily by activated macrophages, monocytes, and dendritic cells in response to infection, tissue injury, or danger-associated molecular patterns. Synthesized as an inactive precursor (pro–IL-1β), it is cleaved into its active form by caspase-1 within multiprotein inflammasome complexes—most notably the NLRP3 inflammasome—and subsequently signals through the IL-1 receptor type 1 (IL-1R1), activating NF-κB and MAPK pathways that drive fever, leukocyte recruitment, endothelial activation, and acute-phase protein production. IL-1β plays a central role in host defense against bacterial, viral, and fungal pathogens, but dysregulated or excessive IL-1β signaling contributes to a wide range of inflammatory and autoimmune conditions, including rheumatoid arthritis, gout, inflammatory bowel disease, type 2 diabetes, atherosclerosis, sepsis, and cytokine release syndromes. Elevated IL-1β is also implicated in neuroinflammatory disorders and certain cancers, where it can promote tumor progression and immune modulation. As both a biomarker and therapeutic target, IL-1β has been the focus of targeted biologic therapies such as IL-1 receptor antagonists and anti–IL-1β monoclonal antibodies. Consequently, human IL-1β is central to research on inflammasome biology, chronic inflammation, metabolic disease, and development of precision anti-inflammatory treatments.
