  • The Ferret/Mink CXCL10 (IP-10) ELISA is designed for the quantitative measurement of CXCL10 (Interferon gamma-induced protein 10, IP-10) in ferret (Mustela putorius furo) and mink (Neovison vison) cell culture supernatants and can also be used to measure CXCL10 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. Ferret and mink CXCL10 proteins are 100% homologous at the amino acid level, enabling the same antibody pair and recombinant standard to be used for accurate detection in both species. CXCL10 is a pro-inflammatory CXC chemokine induced primarily by interferon-γ (IFN-γ) and type I interferons and is produced by monocytes, macrophages, epithelial cells, endothelial cells, and fibroblasts in response to viral infection and immune activation; it functions through the CXCR3 receptor to recruit activated T cells, NK cells, and other immune effector cells to sites of inflammation. In healthy ferrets and mink, circulating CXCL10 levels are typically low or near the limit of detection (often <10-20 pg/mL in serum or plasma), but concentrations can increase to hundreds or thousands of pg/mL during acute viral infections, respiratory disease, systemic inflammation, or experimental immune stimulation, depending on disease severity and timing of sampling. In translational and infectious disease research, particularly in respiratory virus models such as influenza and coronaviruses, CXCL10 serves as a key biomarker of interferon-driven immune responses, cytokine dysregulation, and disease severity. The ELISA kit contains a capture antibody, detection antibody, and recombinant ferret/mink CXCL10 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Ferret (Mustela putorius furo) and Mink (Neovison vison) CXCL10 (C-X-C motif chemokine ligand 10), also known as IP-10 (Interferon gamma–induced protein 10), is a proinflammatory chemokine belonging to the CXC chemokine family, which also includes CXCL9 (MIG) and CXCL11 (I-TAC) that regulate T-cell recruitment during antiviral and inflammatory immune responses. In ferrets and mink, the CXCL10 protein is identical at the amino acid level, indicating strong evolutionary conservation and suggesting similar biological functions in these mustelid species. CXCL10 is produced by macrophages, dendritic cells, epithelial cells, endothelial cells, and fibroblasts in response to interferon-γ (IFN-γ), viral infection, or inflammatory cytokines. It signals primarily through the chemokine receptor CXCR3, which is expressed on activated T lymphocytes, natural killer (NK) cells, and other immune cells, promoting chemotaxis and activation of these cells at sites of infection or inflammation. Ferrets and mink are widely used as animal models for respiratory viral infections, including influenza viruses and SARS-CoV-2, where CXCL10 expression is associated with interferon-driven immune responses and recruitment of antiviral T cells to lung tissues. Because disease progression in these species closely resembles aspects of human respiratory viral infection, characterization of CXCL10 supports research on host–pathogen interactions, immune-mediated lung inflammation, and evaluation of antiviral therapies and vaccines.
