  • The Ovine IFN-γ (Interferon gamma) ELISA is designed for the quantitative measurement of interferon gamma (IFN-γ) in ovine (sheep) cell culture supernatants and can also be used to measure IFN-γ in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IFN-γ is a type II interferon produced primarily by activated CD4⁺ Th1 cells, CD8⁺ cytotoxic T lymphocytes, and natural killer (NK) cells, and it plays a central role in cell-mediated immunity by promoting macrophage activation, enhancing antigen presentation through upregulation of MHC class I and II molecules, and driving Th1-polarized immune responses against intracellular pathogens. In healthy sheep, circulating IFN-γ levels are typically low or undetectable (often <5-10 pg/mL in serum or plasma), but concentrations can increase to tens or hundreds of pg/mL following infection with intracellular bacteria or viruses, vaccination, or mitogen/antigen-specific stimulation, depending on immune status and experimental conditions. In veterinary and translational research, ovine IFN-γ serves as a key biomarker of Th1 immune activation, vaccine efficacy, host-pathogen interactions, and diseases such as mycobacterial infections and viral respiratory disease in large-animal models. The ELISA kit contains a capture antibody, detection antibody, and recombinant ovine IFN-γ protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Ovine Interferon-γ (IFN-γ) is a type II interferon cytokine that plays a central role in cell-mediated immunity and regulation of antimicrobial and inflammatory responses in sheep (Ovis aries). IFN-γ is produced primarily by activated T helper 1 (Th1) cells, cytotoxic T lymphocytes, and natural killer (NK) cells following stimulation by intracellular pathogens or immune signaling molecules such as IL-12 and IL-18. Ovine IFN-γ binds to the interferon-γ receptor (IFNGR1/IFNGR2) expressed on many immune and non-immune cells, activating intracellular signaling through the JAK–STAT pathway, particularly STAT1, which induces the expression of numerous interferon-stimulated genes involved in antigen presentation, macrophage activation, and antimicrobial defense. In sheep, IFN-γ is critical for activation of macrophages, enhancement of MHC class I and II expression, and control of intracellular pathogens including viruses, bacteria, and parasites. Ovine IFN-γ is widely studied in veterinary immunology and livestock disease research; for example, IFN-γ responses are commonly measured in studies of Mycobacterium avium subsp. paratuberculosis infection (Johne’s disease), where IFN-γ production by antigen-stimulated lymphocytes is used as an indicator of cell-mediated immune responses and early infection in sheep.
