  • The Bovine CXCL11 (I-TAC) ELISA is designed for the quantitative measurement of C-X-C motif chemokine ligand 11 (CXCL11) in bovine (cattle) cell culture supernatants and can also be used to measure CXCL11 in additional biological sample types reported in the literature, including serum, plasma, milk, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. CXCL11 is an interferon-inducible CXC chemokine produced primarily by monocytes, macrophages, endothelial cells, and epithelial cells in response to interferon-γ (IFN-γ) and type I interferons, and it functions through the CXCR3 receptor to recruit activated T lymphocytes and natural killer (NK) cells to sites of inflammation, particularly during intracellular infections. In healthy cattle, circulating CXCL11 levels are typically low or near the limit of detection (often <10-20 pg/mL in serum or plasma), but concentrations can increase substantially during viral or bacterial infections, respiratory disease, mastitis, or experimental immune stimulation, depending on disease severity and timing of sampling. In veterinary and translational research, bovine CXCL11 serves as an important biomarker of Th1-type immune activation, interferon-driven inflammatory responses, host-pathogen interactions, and vaccine-induced cellular immunity. The ELISA kit contains a capture antibody, detection antibody, and recombinant bovine CXCL11 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Bovine CXCL11 (C-X-C motif chemokine ligand 11), also known as I-TAC (Interferon-inducible T-cell alpha chemoattractant), is a proinflammatory chemokine belonging to the CXC chemokine family, which also includes CXCL9 (MIG) and CXCL10 (IP-10) that regulate T-cell recruitment during antiviral and inflammatory immune responses. In cattle (Bos taurus), CXCL11 is produced by macrophages, dendritic cells, endothelial cells, epithelial cells, and fibroblasts in response to interferon-γ (IFN-γ), microbial infection, or inflammatory cytokines. CXCL11 signals through the chemokine receptor CXCR3, which is expressed on activated T lymphocytes, natural killer (NK) cells, and other immune cells, promoting chemotaxis and activation of these cells at sites of infection or inflammation. In bovine health, CXCL11 contributes to Th1-type immune responses against intracellular pathogens, including Mycobacterium bovis, the causative agent of bovine tuberculosis, where CXCL11-mediated recruitment of CXCR3⁺ T cells supports granuloma formation and cellular immune defense. Because bovine tuberculosis shares important immunopathological features with human tuberculosis, cattle are used as comparative animal models for tuberculosis research, and bovine CXCL11 is studied as part of the interferon-driven chemokine network involved in T-cell trafficking, host–pathogen interactions, and development of diagnostic and vaccine strategies.
