  • The Equine GM-CSF (Granulocyte-Macrophage Colony-Stimulating Factor, CSF2) ELISA is designed for the quantitative measurement of GM-CSF in equine (horse) cell culture supernatants and can also be used to measure GM-CSF in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, synovial fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. GM-CSF is a hematopoietic growth factor and pro-inflammatory cytokine produced by activated T cells, macrophages, endothelial cells, fibroblasts, and other stromal cells in response to immune stimulation, and it functions by promoting the proliferation, differentiation, activation, and survival of granulocytes and macrophages from bone marrow progenitors. In horses, GM-CSF plays an important role in innate immune responses, airway inflammation, joint inflammation, and host defense against infectious agents. In healthy equine subjects, circulating GM-CSF levels are typically low or near the limit of detection (often <5-10 pg/mL in serum or plasma), but concentrations may increase during respiratory conditions such as equine asthma (recurrent airway obstruction), bacterial or viral respiratory infections, septic arthritis, wound healing, systemic inflammation, or experimental immune stimulation, depending on disease severity and timing of sampling. In veterinary and translational research, equine GM-CSF serves as an important biomarker of myeloid cell activation, inflammatory airway disease, joint inflammation, and immune modulation in performance and clinical equine medicine. The ELISA kit contains a capture antibody, detection antibody, and recombinant equine GM-CSF protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Equine GM-CSF (Granulocyte–Macrophage Colony-Stimulating Factor, CSF2) is a hematopoietic cytokine belonging to the colony-stimulating factor family, which also includes G-CSF (CSF3) and M-CSF (CSF1) that regulate the development, proliferation, and activation of myeloid immune cells. In horses (Equus caballus), GM-CSF is produced by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and other stromal cells in response to infection, inflammatory cytokines, or immune stimulation. Equine GM-CSF binds to the GM-CSF receptor (CSF2R) expressed on hematopoietic progenitor cells and mature myeloid cells, activating intracellular signaling pathways such as JAK/STAT, MAPK/ERK, and PI3K–AKT, which promote the proliferation and differentiation of granulocytes, macrophages, and dendritic cells, as well as enhance their functional activity in innate and adaptive immune responses. In equine health, GM-CSF contributes to immune defense and inflammatory regulation, particularly through activation of macrophages and neutrophils during infection or tissue injury. Horses are also used as animal models for respiratory inflammatory diseases, and GM-CSF–regulated myeloid responses have been studied in equine models of recurrent airway obstruction (heaves), where activation of macrophages and granulocytes contributes to airway inflammation, providing insights relevant to both equine respiratory disease and human asthma research.
