  • The Ferret IL-2 (Interleukin-2) ELISA is designed for the quantitative measurement of IL-2 in ferret (Mustela putorius furo) cell culture supernatants and can also be used to measure IL-2 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-2 is a key T cell growth factor produced primarily by activated CD4⁺ T lymphocytes and, to a lesser extent, CD8⁺ T cells and dendritic cells, and it plays a central role in T cell proliferation, clonal expansion, regulatory T cell maintenance, and the development of adaptive immune responses. In healthy ferrets, circulating IL-2 levels are typically low or near the limit of detection (often <5-10 pg/mL in serum or plasma), but concentrations can increase to tens or hundreds of pg/mL following viral infection, vaccination, mitogen stimulation, or experimental immune challenge, depending on the timing and magnitude of T cell activation. In translational and infectious disease research, particularly in ferret respiratory virus models such as influenza and coronaviruses, IL-2 serves as an important biomarker of T cell activation, vaccine immunogenicity, and adaptive immune response dynamics. The ELISA kit contains a capture antibody, detection antibody, and recombinant ferret IL-2 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Ferret IL-2 (Interleukin-2) is a central T cell–derived cytokine that regulates lymphocyte proliferation, differentiation, and immune homeostasis in ferrets (Mustela putorius furo). IL-2 is produced primarily by activated CD4⁺ and CD8⁺ T lymphocytes following antigen recognition and costimulatory signaling, and it functions through the IL-2 receptor complex (IL-2Rα/CD25, IL-2Rβ, and the common γ chain) to promote clonal expansion of antigen-specific T cells, enhance cytotoxic T lymphocyte and natural killer (NK) cell activity, and support development and maintenance of regulatory T cells (Tregs). In healthy ferrets, basal IL-2 expression is typically low but increases during infection, vaccination, and immune stimulation. Altered IL-2 responses have been observed in viral infections such as influenza and SARS-CoV-2, as well as in inflammatory and immune-mediated conditions. Ferrets serve as important translational animal models for human respiratory viral diseases, including influenza and COVID-19, due to similarities in lung physiology and viral pathogenesis. In biomedical research, ferret IL-2 is used as a biomarker of T cell activation and cell-mediated immunity, supporting studies of vaccine efficacy, antiviral immunity, and immunopathology in respiratory disease models.
