  • The Ovine/Caprine CCL2 (MCP-1) ELISA is designed for the quantitative measurement of C-C motif chemokine ligand 2 (CCL2), also known as monocyte chemoattractant protein-1 (MCP-1), in sheep (Ovis aries) and goat (Capra hircus) cell culture supernatants and can also be used to measure CCL2 in additional biological sample types reported in the literature, including serum, plasma, milk, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. Ovine and caprine CCL2 proteins are identical at the amino acid sequence level, enabling the same antibody pair and recombinant protein standard to be used for accurate detection in both species. CCL2 is a pro-inflammatory chemokine produced by monocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to inflammatory cytokines, infection, and tissue injury, and it functions primarily through the CCR2 receptor to recruit monocytes, memory T cells, and dendritic cells to sites of inflammation. In healthy sheep and goats, circulating CCL2 levels are typically low or near the limit of detection (often <10-20 pg/mL in serum or plasma), but concentrations can increase to tens or hundreds of pg/mL during bacterial or viral infections, mastitis, parasitic disease, respiratory inflammation, parturition-associated stress, or experimental immune stimulation, depending on disease severity and physiological status. In veterinary and translational research, ovine and caprine CCL2 serves as an important biomarker of monocyte recruitment, innate immune activation, inflammatory disease progression, and vaccine-induced immune responses in small ruminant models. The ELISA kit contains a capture antibody, detection antibody, and recombinant ovine/caprine CCL2 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Ovine (Ovis aries) and Caprine (Capra hircus) CCL2 (C-C motif chemokine ligand 2, also known as monocyte chemoattractant protein-1, MCP-1) are highly conserved chemokines that play a key role in recruitment and activation of monocytes, macrophages, and memory T cells during immune responses in small ruminants. Because the ovine and caprine proteins are identical in sequence, they exhibit the same functional properties and receptor interactions. CCL2 primarily signals through the CCR2 receptor, activating intracellular pathways that promote leukocyte migration, cytokine production, and coordination of innate and adaptive immune responses at sites of infection, inflammation, or tissue injury. In sheep and goats, CCL2 is particularly important in inflammatory and infectious diseases such as mastitis, pneumonia, gastrointestinal parasitic infections, and caseous lymphadenitis, where recruitment of monocytes and macrophages is essential for pathogen control and tissue repair. However, excessive or prolonged CCL2 expression may contribute to chronic inflammation and tissue damage. As a biomarker of monocyte/macrophage recruitment and inflammatory activation, ovine and caprine CCL2 are widely studied in investigations of host–pathogen interactions, vaccine responses, and immune regulation in small ruminant production systems. Characterization of CCL2 in these species supports veterinary research aimed at improving disease resistance, understanding inflammatory disease mechanisms, and enhancing the health and productivity of sheep and goats.
