  • The Cynomolgus Monkey GM-CSF (Granulocyte-Macrophage Colony-Stimulating Factor, CSF2) ELISA is designed for the quantitative measurement of GM-CSF in cell culture supernatants from Cynomolgus monkeys (Macaca fascicularis) and can also be used to measure GM-CSF in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. Cynomolgus and Rhesus monkey (Macaca mulatta) GM-CSF proteins are identical at the amino acid sequence level, allowing this assay to be used for accurate detection in both species. GM-CSF is a hematopoietic growth factor and pro-inflammatory cytokine produced by activated T cells, macrophages, endothelial cells, fibroblasts, and other stromal cells in response to immune stimulation, and it functions by promoting the proliferation, differentiation, activation, and survival of granulocyte and macrophage progenitors as well as enhancing the functional activity of mature myeloid cells. In healthy non-human primates, circulating GM-CSF levels are typically low or near the limit of detection (often <5-10 pg/mL in serum or plasma), but concentrations may increase to tens or hundreds of pg/mL during acute inflammation, infection, cytokine release, toxicology studies, vaccine responses, or biologic drug administration, depending on study design and timing of sampling. In non-human primate research, GM-CSF serves as an important biomarker of myeloid cell activation, innate immune responses, cytokine release syndromes, and immunomodulatory drug effects in preclinical safety and efficacy studies. The ELISA kit contains a capture antibody, detection antibody, and recombinant Cynomolgus monkey GM-CSF protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Cynomolgus Monkey GM-CSF (Granulocyte–Macrophage Colony-Stimulating Factor, CSF2) is a hematopoietic cytokine belonging to the colony-stimulating factor family, which also includes G-CSF (CSF3) and M-CSF (CSF1) that regulate the development, proliferation, and activation of myeloid immune cells. In cynomolgus monkeys (Macaca fascicularis), GM-CSF is produced by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to infection, inflammatory cytokines, or immune stimulation. Cynomolgus monkey GM-CSF binds to the GM-CSF receptor (CSF2R) expressed on hematopoietic progenitor cells and mature myeloid cells, activating intracellular signaling pathways such as JAK/STAT, MAPK/ERK, and PI3K–AKT, which promote the proliferation and differentiation of granulocytes, macrophages, and dendritic cells and enhance their immune effector and antigen-presenting functions. Because cynomolgus monkeys share strong genetic and immunological similarities with humans, they are widely used as preclinical animal models for immunology, infectious disease, and vaccine development. GM-CSF–mediated immune responses have been studied in nonhuman primate vaccine models, including SARS-CoV and other viral vaccine studies, where GM-CSF is used as an immune adjuvant to enhance dendritic cell activation and antigen presentation, providing insights into immune mechanisms and vaccine efficacy relevant to human disease.
