  • The Mouse LIF (Leukemia Inhibitory Factor) ELISA is designed for the quantitative measurement of leukemia inhibitory factor (LIF) in mouse (Mus musculus) cell culture supernatants and can also be used to measure LIF in additional biological sample types reported in the literature, including serum, plasma, uterine lavage fluid, cerebrospinal fluid (CSF), and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. LIF is a pleiotropic cytokine belonging to the IL-6 family and signals through the LIF receptor (LIFR) in complex with gp130 to regulate cell survival, differentiation, and proliferation. It is produced by fibroblasts, stromal cells, macrophages, epithelial cells, and other cell types in response to inflammatory stimuli and developmental cues. In mice, LIF plays critical roles in embryonic implantation, maintenance of pluripotency in embryonic stem cells, hematopoiesis, neural development, tissue repair, and modulation of inflammatory responses. In healthy mice, circulating LIF levels are typically low or near the limit of detection (often <10-20 pg/mL in serum or plasma), but concentrations can increase during inflammation, tissue injury, infection, tumor progression, or hormonally regulated reproductive events, depending on physiological and experimental conditions. In basic and translational research, mouse LIF serves as an important biomarker in studies of stem cell biology, reproductive biology, neurobiology, cancer, and immune regulation. The ELISA kit contains a capture antibody, detection antibody, and recombinant mouse LIF protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Mouse Leukemia Inhibitory Factor (LIF) is a multifunctional cytokine belonging to the IL-6 family—which includes IL-6, IL-11, oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), cardiotrophin-like cytokine factor 1 (CLCF1), and IL-27—and signals through receptor complexes containing the shared gp130 subunit. In mice (Mus musculus), LIF binds to the LIF receptor (LIFR) in association with gp130, activating JAK/STAT3, MAPK, and PI3K signaling pathways that regulate cell survival, differentiation, immune modulation, and tissue repair. Murine LIF is best known for its essential role in maintaining pluripotency of embryonic stem cells in vitro via STAT3 activation and for its requirement in embryo implantation and uterine receptivity in vivo. Beyond reproductive and developmental biology, mouse LIF is involved in hematopoiesis, neural regeneration, and modulation of inflammatory responses in models of colitis, lung injury, neuroinflammation, fibrosis, and cancer. Dysregulated LIF/STAT3 signaling has been linked to tumor progression and stromal remodeling in murine cancer models. Because mice are the primary genetic model organism for biomedical research, characterization of LIF has been instrumental in defining IL-6 family signaling mechanisms, stem cell biology, cytokine-driven inflammation, and development of STAT3-targeted therapeutic strategies relevant to human disease.
