  • The Rat LIF (Leukemia Inhibitory Factor) ELISA is designed for the quantitative measurement of leukemia inhibitory factor (LIF) in rat (Rattus norvegicus) cell culture supernatants and can also be used to measure LIF in additional biological sample types reported in the literature, including serum, plasma, cerebrospinal fluid (CSF), uterine lavage fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. LIF is a pleiotropic cytokine belonging to the IL-6 family that signals through the LIF receptor (LIFR) in complex with gp130 to regulate cell survival, differentiation, and proliferation. It is produced by fibroblasts, stromal cells, macrophages, epithelial cells, and other cell types in response to inflammatory stimuli and developmental signals. In rats, LIF plays important roles in embryonic implantation, hematopoiesis, neural development, tissue regeneration, and modulation of inflammatory responses. In healthy rats, circulating LIF levels are typically low or near the limit of detection (often <10-20 pg/mL in serum or plasma), but concentrations can increase during inflammation, tissue injury, infection, tumor growth, or hormonally regulated reproductive processes, depending on physiological and experimental conditions. In basic and translational research, rat LIF serves as an important biomarker in studies of reproductive biology, neurobiology, stem cell regulation, cancer research, and immune modulation. The ELISA kit contains a capture antibody, detection antibody, and recombinant rat LIF protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Rat Leukemia Inhibitory Factor (LIF) is a multifunctional cytokine belonging to the IL-6 family—which includes IL-6, IL-11, oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), cardiotrophin-like cytokine factor 1 (CLCF1), and IL-27—and signals through receptor complexes containing the shared gp130 subunit. In rats (Rattus norvegicus), LIF binds to the LIF receptor (LIFR) in association with gp130, activating JAK/STAT3, MAPK, and PI3K signaling pathways that regulate cell survival, differentiation, immune modulation, and tissue repair. Rat LIF plays important roles in reproductive biology, particularly in embryo implantation and uterine function, as well as in hematopoiesis and neural development. In experimental models, LIF-mediated STAT3 signaling is involved in neuroprotection following brain injury, modulation of inflammatory responses in colitis and lung injury, cardiac remodeling after ischemia, and regulation of tumor growth and fibrosis. Dysregulated LIF expression may contribute to chronic inflammation or pathological tissue remodeling through sustained STAT3 activation. Because rats are widely used in neuroscience, cardiovascular, toxicology, and pharmacology research, characterization of rat LIF supports investigations into IL-6 family cytokine signaling, regenerative mechanisms, inflammatory disease pathways, and development of STAT3-targeted therapeutic strategies relevant to human medicine.
