  • The Zebrafish CCL3 (C-C motif chemokine ligand 3, also known as MIP-1α-like) ELISA is designed for the quantitative measurement of CCL3 in zebrafish (Danio rerio) cell culture supernatants and can also be used to measure CCL3 in additional biological sample types reported in the literature, including whole-body lysates, tissue homogenates, and embryo or larval extracts, with minor protocol modifications recommended to optimize performance across different matrices. CCL3 is a pro-inflammatory chemokine produced primarily by macrophages, neutrophils, and other innate immune cells in response to infection, tissue injury, and inflammatory stimuli, and it functions through CC chemokine receptors to promote leukocyte recruitment and activation at sites of inflammation. In zebrafish, CCL3 plays an important role in innate immune responses during early development and is involved in host defense against bacterial, viral, and parasitic pathogens, as well as in models of tissue injury and regeneration. Basal CCL3 expression in healthy zebrafish is typically low, but levels increase significantly following microbial challenge, chemical-induced inflammation, wounding, or genetic manipulation of immune pathways, depending on developmental stage and experimental conditions. In developmental biology and infectious disease research, zebrafish CCL3 serves as an important biomarker of macrophage recruitment, neutrophil trafficking, and inflammatory signaling in this widely used vertebrate model system. The ELISA kit contains a capture antibody, detection antibody, and recombinant zebrafish CCL3 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Zebrafish (Danio rerio) CCL3 (C-C motif chemokine ligand 3, also referred to as an MIP-1α–like chemokine in teleost fish) is a pro-inflammatory chemokine involved in recruitment and activation of immune cells during innate immune responses. Similar to mammalian CCL3, zebrafish CCL3-like chemokines function to attract macrophages, neutrophils, and other leukocytes to sites of infection, tissue injury, or inflammatory stimulation through chemokine receptor pathways related to CCR1/CCR5-like receptors in fish. In zebrafish, CCL3 expression is induced in macrophages, epithelial cells, and other immune-responsive tissues following exposure to pathogens, inflammatory cytokines, or tissue damage. The chemokine plays an important role in early innate immune responses, particularly during bacterial infections such as Mycobacterium marinum and other experimental infection models where macrophage recruitment and granuloma formation are key components of host defense. Because zebrafish larvae are transparent and genetically tractable, CCL3-mediated leukocyte recruitment can be visualized in real time using fluorescent reporter lines, making zebrafish a powerful model for studying chemokine-driven immune cell migration. Characterization of zebrafish CCL3-like signaling contributes to understanding of evolutionarily conserved chemokine networks, innate immune cell trafficking, and host–pathogen interactions, providing insights relevant to vertebrate immunology and inflammatory disease mechanisms.
