  • The Ferret CCL8 (C-C motif chemokine ligand 8, also known as MCP-2) ELISA is designed for the quantitative measurement of CCL8 in ferret (Mustela putorius furo) cell culture supernatants and can also be used to measure CCL8 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. CCL8 is a pro-inflammatory CC chemokine produced by monocytes, macrophages, epithelial cells, endothelial cells, and fibroblasts in response to inflammatory cytokines, viral infection, and tissue injury, and it functions through chemokine receptors such as CCR1, CCR2, CCR3, and CCR5 to recruit monocytes, T lymphocytes, NK cells, and other immune effector cells to sites of inflammation. In healthy ferrets, circulating CCL8 levels are typically low or near the limit of detection (often <10-20 pg/mL in serum or plasma), but concentrations can increase to tens or hundreds of pg/mL during acute viral infections, respiratory disease, systemic inflammation, or experimental immune stimulation, depending on disease severity and timing of sampling. In translational and infectious disease research, particularly in ferret respiratory virus models such as influenza and coronaviruses, CCL8 serves as an important biomarker of monocyte and lymphocyte recruitment, innate immune activation, and inflammatory disease progression. The ELISA kit contains a capture antibody, detection antibody, and recombinant ferret CCL8 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Ferret CCL8 (C–C motif chemokine ligand 8, also known as MCP-2 – Monocyte Chemoattractant Protein-2) is a proinflammatory chemokine involved in leukocyte recruitment and regulation of inflammatory responses in ferrets (Mustela putorius furo). CCL8 is a member of the CC chemokine family, which includes related chemokines such as CCL2 (MCP-1), CCL7 (MCP-3), and CCL13 (MCP-4) that coordinate monocyte and leukocyte trafficking during immune activation. In ferrets, CCL8 is produced by macrophages, fibroblasts, endothelial cells, epithelial cells, and other immune cells in response to infection, inflammatory cytokines, or tissue injury. CCL8 exerts its biological effects through interaction with chemokine receptors including CCR1, CCR2, CCR3, and CCR5, promoting chemotaxis and activation of monocytes, macrophages, dendritic cells, T lymphocytes, and eosinophils at sites of infection or inflammation. Ferrets are widely used as animal models for respiratory viral infections, particularly influenza and coronaviruses, and chemokines such as CCL8 are important for recruiting immune cells to the respiratory tract and regulating inflammatory responses during viral infection. Consequently, ferret CCL8 is studied in infectious disease, immunology, and vaccine research, helping to elucidate mechanisms of chemokine-mediated immune cell trafficking and host responses to respiratory pathogens.
