  • The Ferret IL-8 (CXCL8) ELISA is designed for the quantitative measurement of interleukin-8 (IL-8/CXCL8) in ferret (Mustela putorius furo) cell culture supernatants and can also be used to measure IL-8 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-8 is a pro-inflammatory CXC chemokine produced by monocytes, macrophages, endothelial cells, epithelial cells, and other cell types in response to inflammatory cytokines, microbial products, and cellular stress, and it functions primarily through CXCR1 and CXCR2 receptors to recruit and activate neutrophils at sites of infection or tissue injury. In healthy ferrets, IL-8 levels are typically low or near the limit of detection in circulation (often <10-20 pg/mL in serum or plasma), but concentrations can increase to tens, hundreds, or higher pg/mL during acute respiratory viral infection, bacterial co-infection, airway inflammation, or experimental immune stimulation, depending on disease severity and timing of sampling. In translational and infectious disease research, particularly in ferret respiratory virus models such as influenza and coronaviruses, ferret IL-8 serves as an important biomarker of neutrophil trafficking, innate immune activation, airway inflammation, and cytokine-driven disease progression. The ELISA kit contains a capture antibody, detection antibody, and recombinant ferret IL-8 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Ferret Interleukin-8 (IL-8), also known as CXCL8, is a potent pro-inflammatory chemokine that plays a central role in innate immune responses in ferrets (Mustela putorius furo) by mediating neutrophil chemotaxis, activation, and recruitment to sites of infection or tissue injury. Produced by respiratory epithelial cells, macrophages, and endothelial cells in response to viral and bacterial stimuli, ferret IL-8 signals through CXCR1 and CXCR2 receptors to promote neutrophil migration and amplification of local inflammatory responses. IL-8 is strongly upregulated during respiratory infections such as influenza virus and SARS-CoV-2, where increased expression correlates with lung inflammation, airway pathology, and disease severity. Because ferrets are widely used as translational models for human respiratory diseases—including influenza, COVID-19, respiratory syncytial virus (RSV), and other airborne pathogens—characterization of IL-8 is critical for understanding neutrophil-driven immunopathology, cytokine and chemokine cascades, and host responses to vaccination or antiviral therapies.
