  • The Swine IL-2 (Interleukin-2) ELISA is designed for the quantitative measurement of IL-2 in porcine (swine) cell culture supernatants and can also be used to measure IL-2 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, lymph node extracts, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-2 is a key T cell growth factor produced primarily by activated CD4⁺ T lymphocytes and, to a lesser extent, CD8⁺ T cells and dendritic cells, and it plays a central role in T cell proliferation, clonal expansion, regulatory T cell maintenance, and the development of adaptive immune responses. In healthy swine, circulating IL-2 levels are typically low or near the limit of detection (often <5-10 pg/mL in serum or plasma), but concentrations can increase to tens or hundreds of pg/mL following viral or bacterial infection, vaccination, mitogen stimulation, or experimental immune challenge, depending on the timing and magnitude of T cell activation. In veterinary and translational research, swine IL-2 serves as an important biomarker of T cell activation, vaccine immunogenicity, cell-mediated immunity, and host-pathogen interactions in porcine disease models such as porcine reproductive and respiratory syndrome virus (PRRSV), swine influenza virus (SIV), African swine fever virus (ASFV), and Mycoplasma hyopneumoniae infection. The ELISA kit contains a capture antibody, detection antibody, and recombinant swine IL-2 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Swine IL-2 (Interleukin-2) is a central T cell–derived cytokine that regulates lymphocyte proliferation, differentiation, and immune homeostasis in pigs (Sus scrofa). IL-2 is produced primarily by activated CD4⁺ and CD8⁺ T lymphocytes following antigen recognition and costimulatory signaling. It functions through the IL-2 receptor complex (IL-2Rα/CD25, IL-2Rβ/CD122, and the common γ chain/CD132) to promote clonal expansion of antigen-specific T cells, enhance cytotoxic T lymphocyte and natural killer (NK) cell activity, and support the development and maintenance of regulatory T cells (Tregs), which are essential for immune balance. In healthy swine, basal IL-2 expression is generally low but increases during infection, vaccination, and immune stimulation. Altered IL-2 responses have been associated with porcine reproductive and respiratory syndrome virus (PRRSV), swine influenza virus, African swine fever virus (ASFV), enteric infections, and immune-mediated inflammatory conditions. Pigs serve as important translational models for human immunology, transplantation research, and vaccine development due to similarities in immune system structure and function. In veterinary and biomedical research, swine IL-2 is widely used as a biomarker of T cell activation and cell-mediated immunity and as a tool for evaluating vaccine efficacy, host–pathogen interactions, and immunomodulatory therapies.
