  • The Black and Large Flying Fox Bat CXCL10 (Interferon gamma-induced protein 10, IP-10) ELISA is designed for the quantitative measurement of CXCL10 in cell culture supernatants from Black Flying Fox (Pteropus alecto) and Large Flying Fox (Pteropus vampyrus) bats and can also be used to measure CXCL10 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. The CXCL10 protein sequence in Black and Large Flying Fox bats is identical at the amino acid level, enabling the same antibody pair and recombinant protein standard to be used for accurate detection in both species. CXCL10 is an interferon-inducible CXC chemokine produced primarily by monocytes, macrophages, endothelial cells, and epithelial cells in response to interferon-γ (IFN-γ) and type I interferons, and it functions through the CXCR3 receptor to recruit activated T lymphocytes and natural killer (NK) cells to sites of inflammation. In healthy bats, circulating CXCL10 levels are typically low or near the limit of detection, but concentrations can increase substantially during viral infection or experimental immune stimulation, depending on pathogen exposure and timing of sampling. In bat immunology and emerging infectious disease research, particularly in studies of zoonotic viruses such as henipaviruses, filoviruses, and coronaviruses, CXCL10 serves as an important biomarker of interferon-driven immune responses, T cell trafficking, and host-pathogen interactions in these natural reservoir species. The ELISA kit contains a capture antibody, detection antibody, and recombinant Black/Large Flying Fox bat CXCL10 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Flying Fox Bat (Pteropus species) CXCL10 (C-X-C motif chemokine ligand 10), also known as IP-10 (Interferon gamma–induced protein 10), is a proinflammatory chemokine belonging to the CXC chemokine family, which also includes CXCL9 (MIG) and CXCL11 (I-TAC) that regulate T-cell recruitment during antiviral immune responses. In flying fox bats, CXCL10 is produced by macrophages, dendritic cells, endothelial cells, epithelial cells, and other tissue cells in response to interferon-γ (IFN-γ), viral infection, or inflammatory cytokines. CXCL10 primarily signals through the chemokine receptor CXCR3, which is expressed on activated T lymphocytes, natural killer (NK) cells, and other immune cells, promoting chemotaxis and activation of these cells at sites of infection or inflammation. Flying fox bats are natural reservoirs for several zoonotic viruses, including Hendra virus and Nipah virus, and CXCL10 likely contributes to coordinating antiviral immune responses and recruitment of cytotoxic immune cells during infection. Studying CXCL10 in flying fox bats helps researchers understand bat antiviral immunity and host–pathogen interactions, including mechanisms that allow bats to control viral infections while limiting excessive inflammatory pathology, providing insights relevant to emerging infectious disease and zoonotic virus research.
