  • The Common Vampire Bat IL-22 ELISA is designed for the quantitative measurement of interleukin-22 (IL-22) in cell culture supernatants from Common Vampire Bats (Desmodus rotundus) and can also be used to measure IL-22 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, intestinal lavage fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-22 is a barrier-protective cytokine produced primarily by activated Th17 and Th22 cells, innate lymphoid cells (ILC3), and certain NK cell subsets, and it signals through the IL-22 receptor complex (IL-22R1 and IL-10R2) expressed predominantly on epithelial and stromal cells to promote antimicrobial peptide production, epithelial integrity, tissue repair, and mucosal defense. In healthy vampire bats, circulating IL-22 levels are typically low or near the limit of detection, but concentrations may increase during viral or bacterial infection, mucosal inflammation, tissue injury, or experimental immune stimulation, depending on pathogen exposure and timing of sampling. In bat immunology and emerging infectious disease research, IL-22 serves as an important biomarker of mucosal immune activation, epithelial barrier responses, and host-pathogen interactions, particularly in studies of zoonotic viruses such as rabies virus and other bat-associated pathogens. The ELISA kit contains a capture antibody, detection antibody, and recombinant Common Vampire Bat IL-22 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Common vampire bat IL-22 (Interleukin-22) is a member of the IL-10 cytokine family that plays an important role in mucosal immunity and epithelial barrier protection in the common vampire bat (Desmodus rotundus). IL-22 is produced primarily by Th17 and Th22 CD4⁺ T cells, as well as by innate lymphoid cells (ILC3s), in response to microbial infection and inflammatory stimulation. It signals through the IL-22 receptor complex (IL-22R1 and IL-10R2) expressed mainly on epithelial and stromal cells, activating downstream JAK/STAT pathways—particularly STAT3—to induce production of antimicrobial peptides, promote epithelial cell proliferation, and enhance tissue repair. In healthy vampire bats, basal IL-22 expression is expected to be low but increases during bacterial or fungal exposure and mucosal immune challenge. Given that Desmodus rotundus is a natural reservoir for rabies virus and other zoonotic pathogens, IL-22 is of particular interest for understanding how bats maintain mucosal barrier integrity while tolerating persistent viral exposure with limited pathology. Study of common vampire bat IL-22 provides insight into epithelial immune regulation, host–pathogen balance, and the unique immune adaptations that characterize bat species.
