  • The Swine IL-16 (Interleukin-16) ELISA is designed for the quantitative measurement of IL-16 in porcine (swine) cell culture supernatants and can also be used to measure IL-16 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, intestinal lavage fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-16 is a pro-inflammatory cytokine produced as a precursor protein by CD4⁺ and CD8⁺ T lymphocytes, monocytes, macrophages, dendritic cells, and epithelial cells, and is cleaved to its active form by caspase-3; it functions primarily as a chemoattractant for CD4⁺ T cells through interaction with the CD4 receptor and plays a key role in coordinating immune cell recruitment, modulating T cell activation, and amplifying inflammatory responses. In healthy swine, circulating IL-16 levels are typically low or near the limit of detection, but concentrations may increase during viral or bacterial infections, respiratory disease, enteric inflammation, systemic inflammatory responses, or experimental immune stimulation, depending on disease severity and timing of sampling. Measurement of swine IL-16 provides valuable insight into T cell-mediated immune activation, leukocyte trafficking, and the balance between innate and adaptive immune responses, making it useful for evaluating disease progression, vaccine efficacy, immune dysregulation, and therapeutic intervention in porcine models such as PRRSV, swine influenza virus (SIV), African swine fever virus (ASFV), and enteric infections. The ELISA kit contains a capture antibody, detection antibody, and recombinant swine IL-16 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Swine Interleukin-16 (IL-16) is a proinflammatory cytokine that plays an important role in regulating immune cell recruitment and activation in pigs (Sus scrofa). IL-16 is produced by several cell types, including CD8⁺ T cells, mast cells, eosinophils, and epithelial cells, in response to immune stimulation or inflammatory signals. Swine IL-16 functions primarily as a chemoattractant for CD4⁺ immune cells by binding to the CD4 receptor, promoting migration and activation of T helper cells, monocytes, and dendritic cells to sites of infection or inflammation. Through these actions, IL-16 contributes to coordination of immune responses and modulation of inflammatory signaling pathways. In pigs, IL-16 participates in immune responses against bacterial, viral, and parasitic pathogens and may influence inflammatory processes in tissues such as the respiratory and gastrointestinal tracts. Swine IL-16 is studied in veterinary immunology and infectious disease research, including investigations of immune responses to important swine pathogens such as porcine reproductive and respiratory syndrome virus (PRRSV), African swine fever virus (ASFV), and Salmonella spp., where IL-16–mediated immune cell recruitment contributes to regulation of inflammatory and host defense mechanisms.
