  • The Swine IL-1 beta (IL-1β) ELISA is designed for the quantitative measurement of interleukin-1 beta (IL-1β) in porcine (swine) cell culture supernatants and can also be used to measure IL-1β in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, synovial fluid, intestinal lavage fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-1β is a potent pro-inflammatory cytokine produced primarily by activated monocytes, macrophages, and dendritic cells following inflammasome activation and caspase-1-mediated cleavage of pro-IL-1β, and it plays a central role in fever induction, leukocyte recruitment, endothelial activation, and amplification of inflammatory signaling cascades. In healthy swine, circulating IL-1β levels are typically very low or near the limit of detection (often <5-10 pg/mL in serum or plasma), but concentrations can increase to tens or hundreds of pg/mL during acute viral or bacterial infections, endotoxemia, respiratory disease, enteric inflammation, joint inflammation, or experimental immune stimulation, depending on disease severity and timing of sampling. Measurement of swine IL-1β provides important insight into innate immune activation, inflammasome activity, and the intensity of inflammatory responses, making it a valuable biomarker in porcine disease models such as porcine reproductive and respiratory syndrome virus (PRRSV), African swine fever virus (ASFV), swine influenza virus (SIV), Mycoplasma hyopneumoniae infection, and bacterial sepsis. The ELISA kit contains a capture antibody, detection antibody, and recombinant swine IL-1β protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Swine Interleukin-1 Beta (IL-1β) is a potent pro-inflammatory cytokine produced primarily by activated macrophages, monocytes, and dendritic cells in pigs (Sus scrofa domesticus) in response to infection, tissue injury, or inflammatory stimuli. Synthesized as an inactive precursor (pro–IL-1β), it is cleaved into its active form by caspase-1 within inflammasome complexes—most notably the NLRP3 inflammasome—and subsequently signals through the IL-1 receptor type 1 (IL-1R1), activating NF-κB and MAPK pathways that promote fever, leukocyte recruitment, endothelial activation, and acute-phase protein production. In swine health, IL-1β plays a central role in inflammatory responses associated with economically significant diseases such as porcine reproductive and respiratory syndrome (PRRS), swine influenza, African swine fever, enteric infections, bacterial pneumonia, and sepsis, where excessive IL-1β production can contribute to lung injury, systemic inflammation, and reduced growth performance. As a biomarker of innate immune activation, IL-1β is widely used in vaccine evaluation, disease pathogenesis studies, and immune monitoring in swine production systems. In translational research, pigs serve as valuable large-animal models due to their physiological and immunological similarities to humans, and characterization of swine IL-1β supports studies of inflammasome biology, cytokine-driven pathology, sepsis, trauma, and development of targeted anti-inflammatory therapies relevant to both veterinary and human medicine.
