  • The Human IL-4 (Interleukin-4) ELISA is designed for the quantitative measurement of IL-4 in human cell culture supernatants and can also be used to measure IL-4 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. The human IL-4 protein sequence is identical to that of chimpanzee (Pan troglodytes), allowing this assay to be used for accurate detection in both species. IL-4 is a key Th2-associated cytokine produced primarily by activated CD4⁺ T helper 2 (Th2) cells, mast cells, basophils, and innate lymphoid cells, and it plays a central role in humoral immunity by promoting B cell proliferation, immunoglobulin class switching (including IgE and IgG4), alternative macrophage activation, and suppression of Th1-type inflammatory responses. IL-4 signals through the IL-4 receptor complex (IL-4Rα paired with either the common γ chain or IL-13Rα1), driving type 2 immune polarization and allergic inflammation. In healthy individuals, circulating IL-4 levels are typically very low or near the limit of detection (often <1-5 pg/mL in serum or plasma), but concentrations may increase during allergic diseases such as asthma and atopic dermatitis, helminth infections, autoimmune disorders with Th2 skewing, and certain malignancies, depending on disease activity and immune status. Measurement of human IL-4 provides valuable insight into Th2 immune polarization, allergic inflammation, antibody-mediated immunity, and cytokine balance in clinical and translational research settings. The ELISA kit contains a capture antibody, detection antibody, and recombinant human IL-4 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Human Interleukin-4 (IL-4) is a central Th2-associated cytokine produced primarily by activated CD4⁺ T helper 2 (Th2) cells, group 2 innate lymphoid cells (ILC2s), mast cells, and basophils, where it plays a pivotal role in regulating humoral immunity, allergic inflammation, and alternative macrophage activation. IL-4 signals through two receptor complexes: the type I receptor (IL-4Rα paired with the common gamma chain, γc) and the type II receptor (IL-4Rα paired with IL-13Rα1), activating JAK/STAT6 pathways that drive B-cell proliferation, IgE class switching, mucus production, and suppression of Th1-associated cytokines such as IFN-γ. IL-4 is critically involved in allergic diseases including asthma, atopic dermatitis, allergic rhinitis, and food allergy, where it promotes IgE-mediated hypersensitivity, eosinophilic inflammation, and tissue remodeling. While essential for defense against helminths and for coordinating effective antibody responses, dysregulated IL-4 signaling contributes to chronic allergic inflammation and barrier dysfunction. IL-4 also influences tumor immunity and fibrotic processes through macrophage polarization and stromal interactions. As a major therapeutic target, blockade of IL-4Rα (thereby inhibiting IL-4 and IL-13 signaling) has demonstrated clinical efficacy in multiple type 2 inflammatory diseases. Consequently, human IL-4 is central to research on Th1/Th2 immune balance, allergy pathogenesis, immune regulation, and development of targeted biologic therapies.
