  • The Cynomolgus Monkey IL-4 (Interleukin-4) ELISA is designed for the quantitative measurement of IL-4 in cell culture supernatants from Cynomolgus monkeys (Macaca fascicularis) and can also be used to measure IL-4 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. The Cynomolgus monkey IL-4 protein sequence is identical to that of Rhesus monkey (Macaca mulatta), enabling this assay to be used for accurate detection in both species. IL-4 is a key Th2-associated cytokine produced primarily by activated CD4⁺ T helper 2 (Th2) cells, mast cells, basophils, and innate lymphoid cells, and it plays a central role in humoral immunity by promoting B cell proliferation, immunoglobulin class switching, alternative macrophage activation, and suppression of Th1-type inflammatory responses. IL-4 signals through the IL-4 receptor complex (IL-4Rα paired with the common γ chain or IL-13Rα1), driving type 2 immune polarization and allergic-type inflammation. In healthy non-human primates, circulating IL-4 levels are typically very low or near the limit of detection, but concentrations may increase during allergic responses, parasitic infection, vaccine-induced immune activation, cytokine modulation studies, or immunotoxicology assessments, depending on experimental conditions and immune status. Measurement of Cynomolgus monkey IL-4 provides valuable insight into Th2 immune polarization, antibody-mediated immunity, immune balance, and therapeutic modulation in preclinical safety and efficacy studies. The ELISA kit contains a capture antibody, detection antibody, and recombinant Cynomolgus monkey IL-4 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Cynomolgus Monkey (Macaca fascicularis) Interleukin-4 (IL-4) is a Th2-associated cytokine produced primarily by activated CD4⁺ T helper 2 (Th2) cells, mast cells, basophils, and group 2 innate lymphoid cells, where it plays a central role in regulating humoral immunity, allergic inflammation, and alternative macrophage activation. Highly homologous to human IL-4, cynomolgus IL-4 signals through type I (IL-4Rα/γc) and type II (IL-4Rα/IL-13Rα1) receptor complexes, activating JAK/STAT6 pathways that promote B-cell proliferation, IgE class switching, mucus production, and suppression of Th1-associated cytokines such as IFN-γ. In nonhuman primate research, IL-4 is particularly relevant in models of allergic airway disease, asthma, parasitic infection, and vaccine-induced immune responses, where Th1/Th2 balance influences protective immunity and immunopathology. IL-4 also plays a role in shaping immune responses during viral infections such as dengue, influenza, and SARS-CoV-2, where excessive Th2 polarization may modulate antiviral immunity. Because cynomolgus monkeys are widely used in preclinical testing of biologics targeting IL-4/IL-13 pathways and in translational immunology studies, characterization of IL-4 is critical for evaluating immune polarization, safety of cytokine-modulating therapies, and mechanisms underlying allergic and inflammatory disease. As a key regulator of type 2 immunity, cynomolgus IL-4 provides important translational insight bridging nonhuman primate models and human Th2-mediated disorders.
