  • The Bovine CCL5 (C-C motif chemokine ligand 5, also known as RANTES) ELISA is designed for the quantitative measurement of CCL5 in bovine (cattle) cell culture supernatants and can also be used to measure CCL5 in additional biological sample types reported in the literature, including serum, plasma, milk, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. The bovine CCL5 protein sequence is identical to that of Bubalus bubalis (water buffalo) and Bubalus kerabau (carabao), enabling this assay to be used for accurate detection across these closely related species. CCL5 is a pro-inflammatory CC chemokine produced primarily by activated T lymphocytes, macrophages, platelets, epithelial cells, and endothelial cells in response to viral infection and inflammatory cytokines, and it functions through chemokine receptors such as CCR1, CCR3, and CCR5 to recruit T cells, monocytes, eosinophils, and other leukocytes to sites of inflammation. In healthy cattle and related buffalo species, circulating CCL5 levels are typically low or near the limit of detection (often <10-20 pg/mL in serum or plasma), but concentrations can increase during mastitis, respiratory disease, enteric infections, bovine leukemia virus (BLV) infection, Mycobacterium bovis infection (bovine tuberculosis), and other inflammatory conditions, depending on disease severity and timing of sampling. Measurement of bovine CCL5 provides valuable insight into T cell trafficking, leukocyte recruitment, antiviral and antibacterial immune responses, and inflammatory disease progression in veterinary and translational research settings. The ELISA kit contains a capture antibody, detection antibody, and recombinant bovine CCL5 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Bovine CCL5 (C–C motif chemokine ligand 5, also known as RANTES – Regulated upon Activation, Normal T cell Expressed and Secreted) is a proinflammatory chemokine that plays an important role in immune cell recruitment and inflammatory responses in cattle (Bos taurus). CCL5 belongs to the CC chemokine family, which includes related chemokines such as CCL3 (MIP-1α), CCL4 (MIP-1β), and CCL2 (MCP-1) that regulate leukocyte trafficking during immune activation. In cattle, CCL5 is produced primarily by activated T lymphocytes, macrophages, dendritic cells, platelets, and epithelial cells in response to infection, inflammatory stimuli, or tissue injury. CCL5 exerts its biological effects mainly through binding to the chemokine receptors CCR1, CCR3, and CCR5, triggering intracellular signaling pathways that promote chemotaxis and activation of T cells, monocytes, macrophages, eosinophils, and natural killer (NK) cells at sites of infection or inflammation. In bovine health, CCL5 contributes to immune responses against bacterial and viral pathogens, including those involved in mastitis, respiratory disease complexes, and viral infections such as bovine viral diarrhea virus (BVDV). By promoting recruitment and activation of immune cells, CCL5 helps coordinate local inflammatory responses and antiviral defense mechanisms. Because chemokine-mediated immune cell trafficking is essential for effective host defense, bovine CCL5 is studied in veterinary immunology, infectious disease research, and mastitis biology, contributing to improved understanding of immune regulation and disease resistance in cattle.
