  • The Swine IL-15 (Interleukin-15) ELISA is designed for the quantitative measurement of IL-15 in porcine (swine) cell culture supernatants and can also be used to measure IL-15 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, intestinal lavage fluid, lymphoid tissue extracts, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-15 is a pleiotropic cytokine produced by monocytes, macrophages, dendritic cells, epithelial cells, and other stromal cells, and it plays a critical role in the development, proliferation, and survival of natural killer (NK) cells and memory CD8⁺ T lymphocytes through signaling via the IL-15 receptor complex (IL-15Rα/IL-2Rβ/γc). In swine, IL-15 is central to antiviral immunity, mucosal immune defense, and maintenance of cytotoxic lymphocyte populations. In healthy pigs, circulating IL-15 levels are typically low or near the limit of detection, but concentrations can increase during viral infections, inflammatory responses, or experimental immune stimulation, depending on disease severity and timing of sampling. In veterinary and translational research, swine IL-15 is particularly relevant in porcine disease and comparative medicine models including porcine reproductive and respiratory syndrome virus (PRRSV), African swine fever virus (ASFV), swine influenza virus (SIV), porcine circovirus type 2 (PCV2), and enteric viral infections such as porcine epidemic diarrhea virus (PEDV), as well as vaccine efficacy and cellular immunity studies. Measurement of swine IL-15 provides valuable insight into NK cell activation, cytotoxic T cell responses, antiviral immunity, and immune memory dynamics in porcine research settings. The ELISA kit contains a capture antibody, detection antibody, and recombinant swine IL-15 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Swine Interleukin-15 (IL-15) is a cytokine that plays an important role in regulating innate and adaptive immune responses in pigs (Sus scrofa). IL-15 is produced by a variety of cell types, including monocytes, macrophages, dendritic cells, and epithelial cells, in response to infection or inflammatory stimuli. Swine IL-15 signals through a receptor complex composed of IL-15Rα together with the shared IL-2/15Rβ (CD122) and common γ chain (γc), activating intracellular signaling pathways such as JAK–STAT that promote lymphocyte proliferation, survival, and activation. In pigs, IL-15 is particularly important for the development, maintenance, and activation of natural killer (NK) cells and memory CD8⁺ T cells, supporting cytotoxic immune responses against infected or abnormal cells. It also contributes to regulation of mucosal immunity and inflammatory responses in tissues such as the respiratory and gastrointestinal tracts. In cell culture, recombinant swine IL-15 is commonly used to stimulate proliferation and survival of NK cells and cytotoxic T lymphocytes, maintain memory CD8⁺ T cell populations, and enhance cytotoxic activity in in vitro immune assays.
