  • The Cynomolgus Monkey TNF-α (Tumor Necrosis Factor alpha) ELISA is designed for the quantitative measurement of TNF-α in cell culture supernatants from Cynomolgus monkeys (Macaca fascicularis) and can also be used to measure TNF-α in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, cerebrospinal fluid (CSF), and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. The Cynomolgus monkey TNF-α protein sequence is identical to that of Rhesus monkey (Macaca mulatta), enabling this assay to be used for accurate detection in both species. TNF-α is a potent pro-inflammatory cytokine produced primarily by activated monocytes, macrophages, T lymphocytes, and NK cells in response to infection, endotoxin exposure, and immune stimulation, and it plays a central role in fever induction, endothelial activation, apoptosis, cytokine cascade amplification, and regulation of innate and adaptive immune responses through signaling via TNFR1 (p55) and TNFR2 (p75). In healthy non-human primates, circulating TNF-α levels are typically very low or near the limit of detection, but concentrations can increase significantly during acute infection, systemic inflammation, cytokine release, toxicology studies, vaccine responses, or biologic drug administration, depending on study design and timing of sampling. Measurement of Cynomolgus monkey TNF-α provides valuable insight into inflammatory burden, cytokine release dynamics, immune dysregulation, and therapeutic response in preclinical safety and efficacy studies. The ELISA kit contains a capture antibody, detection antibody, and recombinant Cynomolgus monkey TNF-α protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Cynomolgus Monkey (Macaca fascicularis) Tumor Necrosis Factor Alpha (TNF-α) (also known as TNF or cachectin) is a proinflammatory cytokine that plays a central role in innate immune responses, inflammation, and host defense against pathogens in nonhuman primates. TNF-α is a member of the tumor necrosis factor (TNF) superfamily, which includes related cytokines such as TNF-β (lymphotoxin-α), Fas ligand (FasL), CD40 ligand (CD40L), and TRAIL, molecules involved in immune signaling, apoptosis, and inflammatory regulation. In cynomolgus monkeys, TNF-α is primarily produced by activated macrophages, monocytes, dendritic cells, and T lymphocytes in response to infection, endotoxin exposure, or tissue injury. TNF-α exerts its biological effects by binding to TNF receptors TNFR1 (p55) and TNFR2 (p75), activating intracellular signaling pathways including NF-κB, MAPK, and caspase-mediated pathways, which regulate cytokine production, immune cell activation, apoptosis, and inflammatory responses. Because cynomolgus monkeys are widely used as preclinical models for human infectious diseases, immunology, and inflammatory disorders, TNF-α has been extensively studied in models of tuberculosis, viral infections, autoimmune disease, and sepsis, where dysregulated TNF signaling contributes to inflammation, tissue damage, and immune-mediated pathology. Due to their close genetic and immunological similarity to humans, characterization of cynomolgus monkey TNF-α is important for translational research and evaluation of anti-TNF therapies and other immunomodulatory treatments prior to clinical development.
