  • The Canine GM-CSF (Granulocyte-Macrophage Colony-Stimulating Factor, CSF2) ELISA is designed for the quantitative measurement of GM-CSF in dog (Canis lupus familiaris) cell culture supernatants and can also be used to measure GM-CSF in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, synovial fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. GM-CSF is a hematopoietic growth factor and pro-inflammatory cytokine produced primarily by activated T lymphocytes, macrophages, endothelial cells, and fibroblasts in response to immune stimulation, and it functions by promoting the proliferation, differentiation, activation, and survival of granulocyte and macrophage progenitors, as well as enhancing the functional activity of mature myeloid cells. In healthy dogs, circulating GM-CSF levels are typically low or near the limit of detection, but concentrations may increase during allergic and inflammatory diseases such as atopic dermatitis and chronic bronchitis, immune-mediated disorders including immune-mediated hemolytic anemia (IMHA) and immune-mediated polyarthritis (IMPA), infectious diseases, and neoplastic conditions. In veterinary and comparative medicine research, canine GM-CSF is particularly relevant in naturally occurring disease models such as lymphoma, osteosarcoma, melanoma, and mammary carcinoma (models for human cancer immunotherapy), as well as osteoarthritis and inflammatory airway disease. Measurement of canine GM-CSF provides valuable insight into myeloid cell activation, inflammatory amplification, tumor-associated immune responses, and evaluation of immunomodulatory or regenerative therapies. The ELISA kit contains a capture antibody, detection antibody, and recombinant canine GM-CSF protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Canine GM-CSF (Granulocyte–Macrophage Colony-Stimulating Factor, CSF2) is a hematopoietic cytokine belonging to the colony-stimulating factor family, which also includes G-CSF (CSF3) and M-CSF (CSF1) that regulate the development, proliferation, and activation of myeloid immune cells. In dogs (Canis lupus familiaris), GM-CSF is produced by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and other stromal cells in response to infection, inflammatory cytokines, or immune stimulation. Canine GM-CSF binds to the GM-CSF receptor (CSF2R) expressed on hematopoietic progenitor cells and mature myeloid cells, activating intracellular signaling pathways such as JAK/STAT, MAPK/ERK, and PI3K–AKT, which promote the proliferation and differentiation of granulocytes, macrophages, and dendritic cells, as well as enhance their functional activity in host defense and antigen presentation. In canine health, GM-CSF contributes to innate and adaptive immune responses during infection and inflammation, particularly through regulation of macrophage and neutrophil activity. Dogs are also valuable comparative animal models for human cancer immunology, and GM-CSF has been studied in canine melanoma models, where GM-CSF–expressing tumor vaccines have been used to stimulate dendritic cell activation and anti-tumor immune responses, providing insights relevant to both veterinary oncology and human cancer immunotherapy.
