  • The Guinea Pig IL-6 (Interleukin-6) ELISA is designed for the quantitative measurement of IL-6 in guinea pig (Cavia porcellus) cell culture supernatants and can also be used to measure IL-6 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-6 is a pleiotropic pro-inflammatory cytokine produced by monocytes, macrophages, endothelial cells, fibroblasts, and other cell types in response to infection, tissue injury, and immune stimulation, and it plays a central role in the acute phase response, fever induction, B cell differentiation, T cell activation, and hepatic production of acute phase proteins. In healthy guinea pigs, circulating IL-6 levels are typically low or near the limit of detection, but concentrations can increase significantly during systemic inflammation, bacterial or viral infection, pulmonary inflammation, endotoxemia, or experimental immune challenge, depending on disease severity and timing of sampling. In biomedical and translational research, guinea pig IL-6 is particularly relevant in respiratory disease models (including tuberculosis and viral airway infection), vaccine development studies, asthma and allergic airway inflammation models, and inflammatory pain or sepsis research. Measurement of guinea pig IL-6 provides valuable insight into systemic inflammatory burden, cytokine-driven immune activation, and disease progression in this established small-animal model. The ELISA kit contains a capture antibody, detection antibody, and recombinant guinea pig IL-6 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Guinea Pig Interleukin-6 (IL-6) is a multifunctional pro-inflammatory cytokine and a central member of the IL-6 cytokine family—including IL-6, IL-11, IL-27, IL-31, leukemia inhibitory factor (LIF), oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), and cardiotrophin-like cytokine factor 1 (CLCF1)—which signal through receptor complexes containing the shared gp130 subunit to activate JAK/STAT and MAPK pathways. In guinea pigs (Cavia porcellus), IL-6 plays a critical role in regulating acute-phase responses, fever, leukocyte recruitment, and B- and T-cell differentiation during infection and inflammation. Guinea pigs are widely used as translational models for human infectious diseases, including tuberculosis, influenza, cytomegalovirus, and hemorrhagic fevers, where IL-6 expression correlates with pulmonary inflammation, systemic immune activation, and disease severity.
