  • The Swine CCL8 (C-C motif chemokine ligand 8, also known as MCP-2) ELISA is designed for the quantitative measurement of CCL8 in porcine (swine) cell culture supernatants and can also be used to measure CCL8 in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, synovial fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. CCL8 is a pro-inflammatory CC chemokine produced by monocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to inflammatory cytokines and microbial stimuli, and it functions through chemokine receptors such as CCR1, CCR2, CCR3, and CCR5 to recruit monocytes, T lymphocytes, NK cells, and other immune effector cells to sites of infection or tissue injury. In healthy swine, circulating CCL8 levels are typically low or near the limit of detection, but concentrations can increase during viral and bacterial infections, respiratory disease, enteric inflammation, systemic inflammatory responses, or experimental immune stimulation, depending on disease severity and timing of sampling. Measurement of swine CCL8 provides valuable insight into leukocyte recruitment, innate and adaptive immune activation, inflammatory disease progression, and host-pathogen interactions in porcine disease and translational models, including porcine reproductive and respiratory syndrome virus (PRRSV), African swine fever virus (ASFV), swine influenza virus (SIV), porcine circovirus type 2 (PCV2), Mycoplasma hyopneumoniae infection, porcine epidemic diarrhea virus (PEDV), bacterial sepsis models, and experimental models of acute lung injury and inflammatory bowel disease that parallel human conditions. The ELISA kit contains a capture antibody, detection antibody, and recombinant swine CCL8 protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Swine CCL8 (C–C motif chemokine ligand 8, also known as MCP-2 – Monocyte Chemoattractant Protein-2) is a proinflammatory chemokine that regulates leukocyte recruitment and inflammatory responses in pigs (Sus scrofa). CCL8 is a member of the CC chemokine family, which includes related chemokines such as CCL2 (MCP-1), CCL7 (MCP-3), and CCL13 (MCP-4) that coordinate monocyte and leukocyte trafficking during immune activation. In swine, CCL8 is produced by macrophages, fibroblasts, endothelial cells, epithelial cells, and other immune cells in response to infection, inflammatory cytokines, or tissue injury. CCL8 exerts its biological effects through interaction with chemokine receptors including CCR1, CCR2, CCR3, and CCR5, promoting chemotaxis and activation of monocytes, macrophages, dendritic cells, T lymphocytes, eosinophils, and other leukocytes at sites of infection or inflammation. In swine health, CCL8 contributes to immune responses against viral and bacterial pathogens, including porcine reproductive and respiratory syndrome virus (PRRSV), swine influenza virus, and bacterial respiratory infections, where chemokine-driven recruitment of immune cells is essential for coordinating antiviral and inflammatory responses. Because pigs share many physiological and immunological similarities with humans, swine CCL8 is also studied in comparative immunology and translational animal models, particularly for research on respiratory infections, inflammatory diseases, and chemokine-mediated immune regulation.
