  • The Swine GM-CSF (Granulocyte-Macrophage Colony-Stimulating Factor, CSF2) ELISA is designed for the quantitative measurement of GM-CSF in porcine (swine) cell culture supernatants and can also be used to measure GM-CSF in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, synovial fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. GM-CSF is a hematopoietic growth factor and pro-inflammatory cytokine produced primarily by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to immune stimulation, and it functions by promoting the proliferation, differentiation, activation, and survival of granulocyte and macrophage progenitors, as well as enhancing the functional activity of mature myeloid cells. In healthy swine, circulating GM-CSF levels are typically low or near the limit of detection, but concentrations may increase during viral or bacterial infections, respiratory disease, systemic inflammation, endotoxemia, or experimental immune challenge, depending on disease severity and timing of sampling. In veterinary and translational research, swine GM-CSF is particularly relevant in porcine disease and comparative medicine models including porcine reproductive and respiratory syndrome virus (PRRSV), African swine fever virus (ASFV), swine influenza virus (SIV), Mycoplasma hyopneumoniae infection, bacterial sepsis models, and experimental models of acute lung injury and inflammatory arthritis that parallel human inflammatory diseases. Measurement of swine GM-CSF provides valuable insight into myeloid cell expansion, inflammatory amplification, cytokine network dynamics, and evaluation of immunomodulatory or vaccine responses in porcine research settings. The ELISA kit contains a capture antibody, detection antibody, and recombinant swine GM-CSF protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Swine GM-CSF (Granulocyte–Macrophage Colony-Stimulating Factor, CSF2) is a hematopoietic cytokine belonging to the colony-stimulating factor family, which also includes G-CSF (CSF3) and M-CSF (CSF1) that regulate the development, proliferation, and activation of myeloid immune cells. In pigs (Sus scrofa), GM-CSF is produced by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to infection, inflammatory cytokines, or immune stimulation. Swine GM-CSF binds to the GM-CSF receptor (CSF2R) expressed on hematopoietic progenitor cells and mature myeloid cells, activating intracellular signaling pathways such as JAK/STAT, MAPK/ERK, and PI3K–AKT, which promote the proliferation and differentiation of granulocytes, macrophages, and dendritic cells, while enhancing their phagocytic activity and antigen-presenting capacity. In swine immunity, GM-CSF contributes to regulation of innate and adaptive immune responses, particularly during viral and bacterial infections affecting the respiratory and gastrointestinal systems. Pigs are widely used as translational animal models for human infectious disease and vaccine research, and GM-CSF–mediated immune responses have been studied in porcine models of influenza infection, where activation of macrophages and dendritic cells helps coordinate antiviral immunity and inflammatory responses, providing insights relevant to both swine health and human respiratory disease research.
