  • The Human IFN-α (Interferon alpha) ELISA is designed for the quantitative measurement of IFN-α in human cell culture supernatants and can also be used to measure IFN-α in additional biological sample types reported in the literature, including serum, plasma, cerebrospinal fluid (CSF), bronchoalveolar lavage (BAL) fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IFN-α is a type I interferon produced primarily by plasmacytoid dendritic cells and virus-infected cells in response to viral nucleic acids and innate immune signaling, and it functions through the IFN-α/β receptor (IFNAR1/IFNAR2) to induce expression of interferon-stimulated genes (ISGs) that mediate antiviral defense, enhance antigen presentation, and modulate innate and adaptive immune responses. In healthy individuals, circulating IFN-α levels are typically very low or near the limit of detection, but concentrations can increase rapidly and transiently during acute viral infections, autoimmune diseases such as systemic lupus erythematosus (SLE), chronic inflammatory disorders, and in response to certain immunotherapies. Measurement of human IFN-α provides valuable insight into antiviral immunity, type I interferon pathway activation, immune dysregulation, and cytokine-driven disease mechanisms in clinical and translational research settings. The ELISA kit contains a capture antibody, detection antibody, and recombinant human IFN-α protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Human Interferon-Alpha (IFN-α) is a type I interferon family consisting of multiple closely related subtypes (e.g., IFN-α1, IFN-α2, IFN-α8, IFN-α14, among others) produced primarily by plasmacytoid dendritic cells and virus-infected cells, where it plays a central role in early innate antiviral immunity. Upon binding to the type I interferon receptor complex (IFNAR1/IFNAR2), IFN-α activates the JAK/STAT signaling pathway, leading to induction of hundreds of interferon-stimulated genes (ISGs) that inhibit viral replication, enhance antigen presentation, promote natural killer (NK) cell cytotoxicity, and shape adaptive immune responses. Human IFN-α is critical in defense against viral infections such as influenza, hepatitis B and C viruses, HIV, SARS-CoV-2, and other emerging pathogens, and impaired or delayed IFN-α responses are associated with severe viral disease. Conversely, excessive or chronic IFN-α production contributes to autoimmune and inflammatory disorders including systemic lupus erythematosus (SLE), type I interferonopathies, and certain dermatologic conditions. Clinically, recombinant IFN-α has been used as a therapeutic agent in chronic viral hepatitis and specific malignancies such as hairy cell leukemia and melanoma. As both a biomarker and therapeutic target, human IFN-α is central to research on antiviral immunity, immune dysregulation, cancer immunotherapy, and development of interferon-modulating treatments.
