  • The Ovine GM-CSF (Granulocyte-Macrophage Colony-Stimulating Factor, CSF2) ELISA is designed for the quantitative measurement of GM-CSF in sheep (Ovis aries) cell culture supernatants and can also be used to measure GM-CSF in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, milk, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. GM-CSF is a hematopoietic growth factor and pro-inflammatory cytokine produced primarily by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to infection and immune stimulation, and it functions through the GM-CSF receptor to promote proliferation, differentiation, activation, and survival of granulocyte and macrophage progenitors, as well as enhance the antimicrobial and inflammatory activity of mature myeloid cells. In healthy sheep, circulating GM-CSF levels are typically low or near the limit of detection, but concentrations may increase during respiratory infections, parasitic disease, mastitis, systemic inflammation, endotoxemia, or experimental immune challenge, depending on disease severity and timing of sampling. In veterinary and translational research, ovine GM-CSF is particularly relevant in sheep models of lung inflammation and acute respiratory distress syndrome (ARDS), parasitic infections, mastitis, vaccine response studies, and comparative immunology research that parallels human inflammatory and pulmonary diseases. Measurement of ovine GM-CSF provides valuable insight into myeloid cell expansion, inflammatory amplification, innate immune activation, and evaluation of immunomodulatory therapies in small ruminant research settings. The ELISA kit contains a capture antibody, detection antibody, and recombinant ovine GM-CSF protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Ovine GM-CSF (Granulocyte–Macrophage Colony-Stimulating Factor, CSF2) is a hematopoietic cytokine belonging to the colony-stimulating factor family, which also includes G-CSF (CSF3) and M-CSF (CSF1) that regulate the development, proliferation, and activation of myeloid immune cells. In sheep (Ovis aries), GM-CSF is produced by activated T lymphocytes, macrophages, endothelial cells, fibroblasts, and epithelial cells in response to infection, inflammatory cytokines, or immune stimulation. Ovine GM-CSF binds to the GM-CSF receptor (CSF2R) expressed on hematopoietic progenitor cells and mature myeloid cells, activating intracellular signaling pathways such as JAK/STAT, MAPK/ERK, and PI3K–AKT, which promote the proliferation and differentiation of granulocytes, macrophages, and dendritic cells, while enhancing their phagocytic and antigen-presenting functions. In ovine immunity, GM-CSF contributes to host defense against bacterial and parasitic infections, particularly through activation of macrophages and neutrophils in tissues such as the lungs and gastrointestinal tract. Sheep are also used as animal models for studying respiratory and inflammatory diseases, and GM-CSF–regulated myeloid responses have been investigated in ovine models of respiratory syncytial virus (RSV) infection, which closely resemble human infant RSV disease, helping researchers understand immune mechanisms driving viral lung inflammation and evaluate potential therapeutic strategies.
