  • The Mouse IL-1ra (Interleukin-1 Receptor Antagonist, also known as IL-1RN) ELISA is designed for the quantitative measurement of IL-1ra in mouse (Mus musculus) cell culture supernatants and can also be used to measure IL-1ra in additional biological sample types reported in the literature, including serum, plasma, bronchoalveolar lavage (BAL) fluid, synovial fluid, and tissue lysates, with minor protocol modifications recommended to optimize performance across different matrices. IL-1ra is a naturally occurring anti-inflammatory cytokine produced primarily by monocytes, macrophages, neutrophils, and epithelial cells, and it functions by competitively binding to the IL-1 receptor (IL-1R1) without inducing signal transduction, thereby inhibiting the pro-inflammatory effects of IL-1α and IL-1β and regulating the magnitude of inflammatory responses. In healthy mice, circulating IL-1ra levels are typically low but detectable, reflecting its role in maintaining immune homeostasis, and concentrations can increase significantly during infection, endotoxemia, autoimmune inflammation, tissue injury, or experimental immune challenge. In biomedical and translational research, mouse IL-1ra is widely studied in models of rheumatoid arthritis, inflammatory bowel disease, sepsis, metabolic inflammation, neuroinflammation, and cytokine-driven inflammatory disorders. Measurement of mouse IL-1ra provides valuable insight into anti-inflammatory counter-regulation, IL-1 pathway activity, cytokine balance, and evaluation of therapeutic strategies targeting IL-1-mediated disease mechanisms. The ELISA kit contains a capture antibody, detection antibody, and recombinant mouse IL-1ra protein standard validated for use together in an ELISA format; kit components are not supplied in matched quantities and may be purchased separately. Mouse Interleukin-1 Receptor Antagonist (IL-1Ra) is an anti-inflammatory cytokine produced by monocytes, macrophages, neutrophils, and other immune and stromal cells in mice (Mus musculus) that functions as a key endogenous regulator of IL-1–mediated inflammation. IL-1Ra competitively binds to the IL-1 receptor type 1 (IL-1R1) without activating downstream signaling, thereby inhibiting the pro-inflammatory actions of IL-1α and IL-1β and limiting activation of NF-κB and MAPK pathways that drive fever, leukocyte recruitment, and tissue inflammation. Murine IL-1Ra plays a central role in maintaining immune homeostasis in experimental models of rheumatoid arthritis (e.g., collagen-induced arthritis), inflammatory bowel disease, sepsis, gout, neuroinflammation, and metabolic disorders. IL-1Ra–deficient mice spontaneously develop inflammatory polyarthritis, highlighting its critical function in controlling IL-1–driven pathology. As both a biomarker and therapeutic analog, mouse IL-1Ra is widely used in preclinical research to study inflammasome regulation, cytokine network balance, and the efficacy of IL-1–targeted therapies, providing foundational insight into inflammatory mechanisms and treatment strategies relevant to human autoimmune and inflammatory diseases.
